Objective: Observational study of the protein and m RNA expressions of BSEP and NTCP under the drug intervention by establishing the cholecystolithiasis model in mice.This paper expounds the therapeutic effect of da-huanglingxian capsule on preventing and treating cholecystolithiasis,and further explores the formation mechanism of cholecystolithiasis and the intervention effect of da-huanglingxian capsule.Methods: 60 healthy SPF C57BL/6 male mice were randomly divided into 5 groups: general food group(P group),lithogenic diet group(Z group),lithogenic diet + injecting water group(Z+S group),lithogenic diet + injecting da-huanglingxian capsule group(Z+D group),lithogenic diet + injecting ursodeoxycholic acid group(Z+X group),12 mice in each.Mice were fed according to the requirements of each group about eight weeks.On the one hand,the gallbladder tissue of mice was used to observe the modeling.On the other,Western blotting and real time PCR were used to detect and compare the expression of BSEP and NTCP protein and the transcription level of BSEP and NTCP m RNA.Results:(1)P group formation of stones in 0 cases,formation rate 0%,Z group formation of stones in 11 cases,formation rate 91.7%.After statistical test,there is statistically significant difference between P group and Z group(P<0.05),and the modeling is successful.Z+S group formation of stones in 11 cases and 1 case death,formation rate 90.9%,Z+D group formation of stones in 3 cases,formation rate 25%,there is also statistically significant difference between Z+S group and Z+D group(P<0.05).Z+X group formation of stones in 2 cases,formation rate 16.7%.Compared to Z+D group,the difference between Z+D group and Z+X group is not statistically significant(P>0.05).The results showed that da-huanglingxian capsule could prevent the formation of cholecystolithiasis as well as ursodeoxycholic acid.(2)Western Bloting: The protein expression of BSEP and NTCP is different after 2 months,Z+S group <Z+D group =Z+X group,there is statistically significant difference among them(P<0.05).(3)RT-PCR: The transcription level of result is consistent with Western Blotting,Z+S group <Z+D group =Z+X group.there is also statistically significant differences in the transcription level of BSEP and NTCP(P<0.05).Conclusion:(1)Da-huanglingxian capsule can prevent and cure the formation of cholecystolithiasis in the process of making model in mice.(2)Da-huanglingxian capsule could increase the expression of BSEP and NTCP protein and m RNA transcriptional level in the hepatobiliary membrane of the cholecystolithiasis model mice,and the increased level is equivalent to ursodeoxic acid.(3)Da-huanglingxian capsule can enhance the operation of bile acid by transporters on the membrane of hepatobiliary duct and inhibit the formation of gallstones in mice.
