Objective: To study the effects of human leukemia related protein 16(LRP16)on the proliferation and invasion of colon cancer cells by regulating Wnt/?-catenin pathway.Method: CCD841 Co N cells and 5 kinds of colon cancer cells were selected as the research objects,the expression level of LRP16 in different cells was measured by real-time fluorescence quantitative analysis(RT q PCR),HCT116 cells were transfected with si-LRP16,the proliferation,invasion and migration of HCT116 cells were measured by MTT assay,Transwell cell assay and scratch test respectively;and the expression levels of GSK3?,p-GSK3?,?-Catenin,Ki-67,PCNA,E-cadherin and N-cadherin in HCT116 cells were determined by Western blot.Results: There was no significant difference in LRP16 expression level between any two of CCD841 Co N cell group,Lo Vo cell group and HT29 cell group(P > 0.05),compared with the above three groups,the expression level of LRP16 in COLO205 cell group,SW620 cell group and HCT116 cell group increased significantly(P <0.05),and LRP16 was the most expressed in HCT116 cells,so the following experiments were carried out in this cell.After si-LRP16 transfection,compared with the blank control group and si-LRP16 negative control group,the expression levels of LRP16,p-GSK3?,?-catenin,Ki-67,PCNA and N-cadherin in si-LRP16 group decreased significantly(P < 0.05),the expression level of E-cadherin increased significantly(P < 0.05),and the proliferation,invasion and migration of HCT116 cells decreased significantly(P < 0.05).Conclusions: LRP16 may decrease the proliferation,invasion and migration of HCT116 cells by inhibiting Wnt /?-Catenin pathway activity. |
