Objective: The aim of this study was to investigate the effects of overtraining on intestinal mucosa barrier in rats,to probe into the effect of supplement complex plant polysaccharides on Intestinal barrier,and to analyze the effect of polysaccharides on oxidative stress,immunomodulatory,to protect the intestinal barrier integrity,and then to discuss the possible mechanism.Methods:40 male rats were randomly divided into control group(C,n=10),polysaccharides group(P,n=10)overtraining group(E,n=10)and polysaccharides + overtraining group(PE,n=10).The E and PE groups of rats were trained for 4 weeks general training and 4 weeks of exhaustion training,5 times a week,Group C and E were administrated with physiological saline,the E group and PE group were intragastric administration of polysaccharides(0.5g/kg body weight)with the concentration solution of 10% after training.8 weeks later,determination of blood index: RBC,HGB,WBC;Oxidation antioxidant index: T-SOD,T-AOC,MDA,GSH-Px;Motor function index: BUN,CK,Testosterone;Intestinal mucosal barrier integrity indexes: ET,D-LA,DAO.ELISA measured in serum endotoxin as well as the inflammatory cytokine indexes including IL-1?,IL-6,IL-10,TNF-?,IFN-?.Results:(1)Compared with group C,the RBC in group E was decreased(P<0.05),the WBC decreased significantly(P<0.01),and the HGB decreased significantly(P<0.01).Compared with E group,the number of RBC,WBC and HGB increased in PE group(P<0.05).(2)Compared with group C,the CK and BUN in group E was significantly increased(P<0.01),the Testosterone was significantly decreased(P<0.01);compared with group E,the CK and BUN group PE were significantly decreased significantly(P<0.01),the Testosterone value was significantly increased(P<0.01).(3)Compared with group C,the T-SOD,T-AOC,GSH-Px in group E were significantly decreased(P<0.01),MDA was significantly increased;compared with group E,the T-SOD in group PE have no difference(P>0.05),while the T-AOC,GSH-Px was increased(P<0.05),and the MDA value was significantly decreased(P<0.01).(4)Compared with group C,the rat intestinal IL-1?,IL-6,TNF-?in group E were increased significantly(P<0.01),while the IL-10,IFN-? significantly decreased(P<0.01);compared with group E,the intestinal IL-1?,IL-6,TNF-?in group PE significantly decreased(P<0.01),while the TNF-?value was decreased(P<0.05),and the IL-10 was significantly increased(P<0.01),IFN-? was increased(P<0.05).(5)Compared with group C,the ET,DAO and D-LA of group E were increased significantly(P<0.01).Compared with E group,the ET,DAO and D-LA in group PE were also significantly decreased(P<0.01).Conclusion:(1)Overtraining leads to decrease in the body's exercise ability,and the complex plant polysaccharides intervention can significantly improve the body's exercise ability.(2)Overtraining leads to the increase of oxidative stress in the body,and complex plant polysaccharide intervention can significantly improve the antioxidant capacity of the body.(3)Overtraining resulted in the increase of intestinal proinflammatory factors and the decrease of anti-inflammatory factors.The intervention of complex plant polysaccharides can significantly improve the immune regulation ability of the body.(4)Overtraining resulted in increased intestinal permeability and increased risk of bacterial translocation.It seemed that complex plant polysaccharides intervention can prevent bacterial translocation caused by increased intestinal permeability and protect intestinal mucosal barrier integrity.
