Iron is not only an indispensable trace element to maintain the metabolism of cells,but also an important raw material for the synthesis of hemoglobin(Hb),it plays an important role in the body.With the growth of age,the body's organs and functions began to decline,the immune function gradually decreased;In addition,the aging of body will also increase inflammation in the kidney,which will increase the incidence and mortality of various chronic diseases.Some studies have shown that iron storage organs,such as liver and spleen,will appear iron deposition phenonmenon with the growth of age.Previous experimental studies have shown that the renal iron content increased significantly after the middle age and old age,which is easy to cause peroxidative damage.It remains to be futher studies that aerobic exercise can promote the redistribution of iron level in the body,reduce the iron level in the body,and thereby improve the iron deposition in the elderly kidney,alleviate renal peroxidation injury,and prevent chronic kidney disease.In this experiment,adult mice were used as experimental animal models to explore the relationship between renal inflammation and iron metabolism with aging,and the influence of aerobic exercise on renal iron content and the causes,so as to provide experimental basis for the prevention of renal iron overload related diseases by aerotic exercise.Research contents and methods:In this experiment,20 male mice of C57BL/6 strain mice,13-months-old,were randomly divided into control group(C)and exercise group(E)with 10 mice in each group.The mice in the control group were fed quietly,and the mice in the exercise group were intervened by aerobic exercise.The mice in exercise group did treadmill exercise for 1 week,as adaptive training.The formal treadmill exercise was had perfomed for 22 weeks,and the treadmill exercise was 6 days a week,with one day to rest,and the exercise time was 45 minutes each time.The slope of the treadmill was0.The speed increased by 1 m/min every two weeks,and the speed increased from 10m/min to 15 m/min;then decreased by 1 m/min o 10 m/min every two weeks.Material were took after 22 weeks of aerobic exercise intervention.Serum iron,Serum ferritin,Total iron binding capacity and kidney tissue iron were measured to detect the expression of Hepcidin mRNA and the expression of DMT1,FPN1,TfR1 and il-6 proteins in the kidney.All experimental data were analyzed by SPSS 20.0 software for data processing and statistical analysis.The values used were expressed as “mean ± standard deviation”.The comparison between groups was performed by independent sample T-test.P<0.05 indicates that the difference is significant,and P<0.01 indicates that the difference is highly significant.Research results:(1)Effects of aerobic exercise on iron level in mice: Transferrin saturation,serum iron and Total iron binding capacity level in the exercise group were significantly higher than those in the control group(P<0.01),Serum ferritin level were also higher than the control group(P<0.05).After 22 weeks of aerobic exercise intervention,the iron content in the kidney of the exercise group was significantly lower than that of the control group(P<0.01).The iron content in the liver and spleen of iron storage organs was also significantly reduced(P<0.01).The iron content of the heart and bone marrow tissue was lower than that of the control group(P<0.05).The iron content in skeletal muscle tissue was significantly higher than that in the control group(P<0.01).(2)Effects of aerobic exercise on inflammatory factors in mice: After 22 weeks of aerobic exercise intervention,serum IL-6 levels in the serum group of mice were significantly decreased(P<0.01);IL-6 protein expression in exercise group The amount was significantly lower than the control group(P<0.01).(3)Effects of aerobic exercise on Hepcidin mRNA in mice kidney: After 22 weeks of aerobic exercise intervention,serum IL-6 level were significantly decreased(P<0.01).The expression of Hepcidin mRNA in mouse kidney was significantly declining(P<0.01).(4)Effects of aerobic exercise on DMT1,TfR1,and FPN1 proteins in kidney of mice: After 22 weeks of aerobic exercise intervention,the expression levels of DMT1(+IRE),DMT1(-IRE)and FPN1 protein in exercise group were significantlyhigher than those in control group(P<0.01),the expression of TfR1 protein was also higher than that of the control group(P<0.05).Analysis conclusions:(1)With the increase of age,the iron level of the body increases,and iron deposits appear in various organs.After the intervention of aerobic exercise,the iron consumption in the body is accelerated,the iron level of the body is lowered,and the iron overload phenomenon of the kidney is alleviated.(2)Aerobic exercise increases the expression of iron-related proteins TfR1 and DMT1 in the kidney.The increase of iron intake increases the expression of FPN1,which promotes the release of iron,accelerates the recycling of iron,and transports more iron from the kidney to the blood.,thereby improving kidney iron deposition.(3)Aerobic exercise reduced the expression of IL-6 protein and serum IL-6 in the body,and the expression of Hepcidin mRNA in the kidney was also down-regulated,which effectively reduced the renal inflammatory response and slowed down the aging rate of the body.
