Objective:to construct a model of treadmill exercise with diffe rent intensity in rats,and to evaluate the lactic acid concentration of fast and slow muscles and the effect on the expression of muscl e factor IL-15 and IL-15mRNA in rats induced by different exercise modes.This study can enrich the theoretical basis of sports promoti ng health,and provide theoretical reference for the formulation of s ports programs for mass sports.Methods:the experiment was divided into two parts.The first part was an exploratory study on monitoring exercise intensity with lactic acid.Treadmill exercise on the second part is to detect different ways of rat gastrocnemius and soleus IL-15 content and the influence of IL-15mRNA express,through the complete par ticular strength in rats after treadmill exercise tail,blood lactic aci d,lactic acid to test at the same time take the rat(fast)of the ga strocnemius and soleus(slow),respectively,the two types of muscle with ELISA to detect the contents of IL-15,RT-PCR detection of I L-15 mRNA expression.The first part of the object of different inte nsity treadmill model experiment is 24 adult male SD rats,weight i s 150-22g,divided into 5 groups,each group of five number,accor ding to treadmill exercise intensity is divided into high intensity ex ercise group(LHE)for a long time,short time high intensity exerci se group(SHE),intermittent high intensity exercise group(HIE),int ermittent high strength uphill exercise group(HIUE),high intensity exercise group(CHE).The sampling time is within 3 minutes immedi ately after exercise.In the second part,ELISA and rt-pcr were performed on 24 ad ult SD male rats weighing 220±22g,divided into 8 groups with 3 mice in each group.According to the movement pattern is divided in to:low intensity exercise group(LE),medium intensity intermittent movement(MIE),medium intensity exercise group(MCE),continuou s uphill medium intensity exercise group(MCUE),high strength and keep on moving uphill exhaustion group(HCUE)short,high intens ity intermittent movement exhaustion group(HSIE),high intensity in termittent movement(HIE),high strength uphill alternating group(H AUE).The sampling time was immediately after the movement.For da ta statistics,one-way anova and paired sample T test were performe d with SPSS21.0.Results:(1)The content of IL-15 in fast muscle of LE group was signi ficantly higher than that of all other groups(p<0.01).In addition,t he concentration of MIE/HCUE/HIE group was also significantly diff erent from that of other groups(p<0.01).The content of IL-15 in middle and high intensity intermittent exercise was higher,while the content of IL-15 in high intensity continuous uphill exercise group was lower than that in high intensity continuous uphill exercise gro up.The content of IL-15 in slow muscle of LE group was significa ntly higher than that of MIE/MCE/MCUE/HCUE/HSIE/HAUE group,but there was no significant difference between HIE group and HIE group.Middle and low intensity continuous exercise could increase the content of IL-15 in rats.(2)There was no significant difference in the expression of IL-15mRNA between fast muscle and slow muscle and other groups.In fast muscle,the expression of IL-15mRNA was higher under inter mittent exercise of medium and high intensity.In the slow muscle,medium and low intensity continuous exercise can increase the cont ent of IL-15mRNA in rats.(3)The concentration of IL-15 in muscle and slow muscle was significantly different from that in LE and HCUE groups(p<0.05),and MCE,HIE,HAVE was extremely significantly different.Under intermittent exercise,the expression of IL15mRNA in fast muscle w as higher than that in slow muscle,while under continuous exercise,the expression of IL-15mRNA in fast muscle was lower than that in slow muscle.(4)There was no significant difference in the expression of IL-15mRNA between fast muscle and slow muscle.The expression of I L-15mRNA in fast muscle was higher than that in slow muscle und er medium and low intensity exercise,and the expression of IL15m RNA in slow muscle was higher than that in fast muscle under hig h intensity and long time exercise.Conclusion:(1)The exercise model designed in this study is basically medi um and high intensity exercise,and the exercise model is feasible.S hort interval exercise is conducive to the accumulation of lactic aci d.(2)The lactic acid value measured by low-intensity and mediu m-intensity intermittent exercise was higher;The low lactic acid valu e of high intensity prolonged exercise may be caused by the remov al of lactic acid due to the long exercise time,suggesting that the highest lactic acid value can be measured by short interval exercise.(3)In fast muscles,moderate and low intensity intermittent exe rcise can increase the content of IL-15 and the expression of IL-15 mRNA;In the slow muscle,the level of IL-15 and the expression of IL-15mRNA were increased by moderate and low intensity continuo us exercise.It indicated that IL-15 and IL-15mRNA in fast muscle w ere more sensitive to intermittent exercise at medium and low inten sity,while IL-15 and IL-15 mRNA in slow muscle were more sensi tive to continuous exercise.(4)High-intensity intermittent exercise and high-intensity intermi ttent strenuous exercise will result in low IL-15 content and IL15m RNA expression,which are obvious in fast muscles but not in slow muscles.It suggested that the exercise style with high intensity and long duration was not conducive to the production of IL-15.
