Purpose:To investigate the effects of exercise(including AICAR simulation exercise)intervention on skeletal muscle mitochondrial biosynthesis and microcirculation in mice,and the correlation between skeletal muscle mitochondrial biosynthesis and microcirculation in mice.Preliminary studies on the correlation between skeletal muscle mitochondrial biosynthesis and microcirculation provide a theoretical basis for the prevention and treatment of people with metabolic diseases and / or microcirculation dysfunction,reduce the occurrence of body injuries and diseases,and promote body health.Methods:32 6-week-old C57 BL / 6 healthy male mice were randomly divided into 4groups,a quiet control group(group C,n = 8),an AICAR drug activation group(group A,n = 8),and aerobic exercise group(group M,n = 8),AICAR drug activation+ aerobic exercise group(group AM,n = 8),pre-test of microcirculation.Group C did not do any intervention;1 hour before each exercise intervention,AICAR was injected intraperitoneally in groups A and AM at a dose of 250 mg / kg / day.M and AM mice were subjected to treadmill exercise training intervention for 6 weeks,5days / week,once a day.The specific exercise plan is to run at a speed of 10 m / min for 30 minutes in the first week,and to increase the running speed by 2 m / min per week,the exercise time by 10 minutes per week,and the fifth week.The sixth week keeps the same speed and time as the fourth week,16 m / min,60 min.After the6-week intervention experiment,a microcirculation post-test was performed,and then the mice were weighed and sampled,and the neck was killed.Take the fresh gastrocnemius muscle on the left side and fix it with fixative solution for histological observation to observe the immunohistochemical expression of CD31 and VEGF.The fresh gastrocnemius muscle on the right side is quick-frozen with nitrogen and frozen at-80 ? for Western blot)Experiments to detect changes in mitochondrial biosynthetic protein indicators.Results:(1)After the 6-week aerobic exercise intervention,the body weight of the four groups of mice had increased to a different extent than before the intervention.Among them,the weight gain of the group A mice was more obvious,and there was a significant difference compared with before the intervention(P <0.05).(2)Compared with group C mice,the expression levels of gastrocnemius PGC-1?,SIRT1,TFAM,and CYC proteins in the gastrocnemius muscles of group A,M and AM mice were significantly increased(P <0.05);the gastrocnemius AMPK protein of mice in group M and AM The expression level was significantly higher than that ingroup C,and there was a significant difference(P <0.05).The expression level of gastrocnemius muscle P-AMPK protein in AM group was significantly higher than that in group C,and there was a significant difference(P <0.05).(3)The skin blood perfusion volume,heating blood perfusion volume,blood flow reserve capacity,and skeletal muscle blood perfusion volume of mice in the M and AM groups were significantly higher than those before the exercise training,and were significant(P <0.05);Compared with mice,skin blood perfusion volume,heating blood flow perfusion volume,blood flow reserve capacity,and skeletal muscle blood perfusion volume were significantly increased in M and AM groups(P <0.05);skin heating blood flow in mice in group A perfusion volume and blood flow reserve capacity were also significantly higher than before exercise intervention(P <0.05);skeletal muscle blood flow perfusion volume in group A mice was also significantly higher than that in group C mice(P <0.05).(4)The total CD31 and VEGF IOD values in the AM group were significantly higher than those in the C group,and they were extremely significant(P <0.01).The total IOD value of group A mice and the total IOD value of group M mice were significantly higher than those of group C mice(P <0.05).(5)Heated blood flow perfusion volume and blood flow reserve capacity are moderately positively correlated with PGC-1? protein expression(P <0.05).Heated blood flow perfusion volume and skeletal muscle blood flow perfusion volume are lowly positively correlated with TFAM protein(P <0.05).Blood flow reserve capacity and P-AMPK protein showed a low negative correlation(P <0.05),skeletal muscle blood flow perfusion was moderately negatively correlated with CYC(P <0.05),and heated blood flow perfusion was positively correlated with COXIV(P <0.05).Conclusion:(1)6-week aerobic exercise can promote mitochondrial biosynthesis in skeletal muscle of mice,and increase skin microcirculation and skeletal muscle microcirculation in mice.AICAR + aerobic exercise can effectively promote mitochondrial biosynthesis in skeletal muscle of mice.(2)There is a correlation between skeletal muscle mitochondrial biosynthesis and microcirculation.6-week aerobic exercise may increase skeletal muscle microcirculation in mice,which may be related to AMPK-mediated pathways,and may be related to other pathways of mitochondrial synthesis.
