Halophilic(salt-tolerant)bacteria is a kind of microorganism in extreme environments,it not only lives in salt works,salt flats,saline-alkali land,but also abounds in the pickled foods.As halophilic(salt-tolerant)bacteria has the unique physiological characteristics,it has a broad application prospect,but a large number of breeding will also pose a threat to food safety.This study aimed at use the halophilic bacteria isolation and identification in the salt water which cause pickled eggs corrupted.High-throughput sequencing technology was also used to analysis the microbial diversity of the salt water lead to corruption.We isolated 20 strains of halophilic bacteria,they all could growth in 15?~40?,which best growth at 25?~35?,pH is from 6 to10,better at the range of pH 8~10,18 strains of 20 were salt-tolerant bacteria and 2 strains belong to moderate halophilic bacteria.12 strains which can survive more than 15% salt were identified by using physiological and biochemical identification and 16 S rRNA sequencing.The results indicated that there were 7 strains belong to Salinicoccus,1 strain belongs to Microbacterium,1 strain belongs to Proteus,2 strains belong to Salinivibri,2 strains moderate halophilic bacteria could grow well in salt concentration of 5%-20%.High-throughput sequencing discovered that the microbial diversity in the salt water corrupted.Showed higher abundance compared to the uncorrupted.Their relative abundance as follows: Halanaerobium is 39.62%,NB1-n_norank is 15.21%,Halomonas is 3.82% and Thermophagus is 11.99%,Desulfovermiculus is 4.98%.After the primary and secongdary eggs picked experiments by vaccinate halophilic.5 strains were identified as corrorruption strains,3 strains of them belong to Staphylococcus,1 strain belongs to Salinivibrio and 1 strain belongs to Proteus.Also in the high-throughput sequencing,a groups of Halomonas,as conditional pathogenic bacteria was discovered in a large amount in salted egg brine corruption was concluded as bacteria Halomonas and thoose 5 strains.Besides genus of the Halanaerobium,NB1-n_norank and Thermophagus probably were potential factors which cause salted eggs brine corrupted.
