Many emerging infectious diseases(EIDs)have occurred in recent years.approximately 75%of EIDs originate from wildlife directly or via certain intermediate hosts.Rodents are the most diverse species of mammals and are widely distributed in the world.Rodents are in close contact with humans and are the natural reservoir of a variety of emerging viruses that can spread more than 60 known zoonotic infectious diseases,Therefore,a large-scale systematic research was conducted on the virome of rodents in China.This study as a part of the china rodents virome focuses on the anelloviruses.As no efficient cell line model and suitable animal model to sustain viral replication study,the pathogenic potential of anellviruses is still unclear.Some genotypes or genogroups are suggested to be pathogenic and some have been related to the progression of syndromes caused by other viral agents.Recently anelloviruses have been identified in humans and other mammalian hosts.Rodent anelloviruses have previously been reported in the UK.However,there is no report about rodents carrying the anelloviruses in China at present.Therefore,it is very necessary to understand the existence of anelloviruses in various rodents from different regions.In this study,we performed a systematic survey of rodent-borne anelloviruses using virome data from rodents collected in 20 provinces throughout China from sites covered by the National HFRS(hemorrhagic fever with renal syndrome)Surveillance Network..The animals belonged to 42 species in the order Rodentia.2416 pharyngeal and anal swabs samples was examied in this study.The results show that almost all of those rodents were infected with anelloviruses that expand the type of rodent host.Based on the partial gemomic sequences of the virus obtained by the metagenomic analyses,we designed specific nested primers for PCR to screen for each virus in individual samples.Eleven representative complete genomes and two partial genomes were amplified.The RoTTV genome size ranges from 2.3 to 2.6kb.According to the sequence assamble and gene annotation results,the RoTTV genomes we got in this study encode 2-3 open reading frames(ORFs),in addition to a non-translated region(UTR)of about495-868bp.From the phylogenetic analysis of complete ORF1 of thirteen anellovirus strain,our strains form four lineages(1-4)and respectively were suggested to be new genus in the Anelloviridae based on the genomic structure,sequence distance,phylogenetic analysis results.The new RoTTV expands the vital taxa.Our results provided the evidence that the Mus musculus can be infected by the Rodents anellovirus strain RtMm-AneV/ZJ2016.It suggests that the laboratory mice could be infected by strain RtMm-AneV/ZJ2016 and would be used in the study of pathogenicity and viral cycle of anellovirus.We outlined the rodent anellovirus viromes and identified 13 novel anellovirus strains in China.These data also provide a meaningful basis for the identification and tracking of potential rodent-origin anelloviruses.
