Sorex isodon is a small mammal which belongs to the Sorex of the Insectivora(Soricidae).Studied on genetic diversity and molecular phylogenetics of the Sorex isodon in northeastern China.Research on genetic diversity shows:(1)Seventy-seven samples of the Sorex isodon from three populations in the Northeast China(Daxinganling Mountains,Xiaoxinganling Mountains and Changbaishan Mountains)were sequenced.The entire sequence of the obtained Cyt b gene was analyzed,A total of 64 haplotypes were obtained.The complete sequence of Cyt b gene was 1140 bp,and 64 haplotypes were obtained from 77 samples.The haplotype diversity was 0.9920,the average number of nucleotide differences was 11.9490,and the nucleotide diversity was 0.0105.From above,the genetic diversity was high in the northeast of China.From genetic diversity parameters of three populations,the genetic diversity of Changbaishan Mountains population is the highest(h=0.9890?i=12.5810??=0.0110).It is suggested that the Changbaishan Mountains may be the refuge of last glacial for Sorex isodon.(2)Analysis of three populations of Sorex isodon in Northeast China,the range of genetic distance between populations was 0.0062-0.0154.The genetic distance between sampling points is between 0.0039-0.0152.The genetic distances of the three populations of the Sorex isodon in Northeast China were positively correlated with the geographical distribution.(3)The experimental results of F-statistics and genetic similarity coefficients between the populations of Sorex isodon in Northeast China were consistent.Fully consistent with the geographical distance between the populations.(4)Phylogenetic tree shows that the Sorex isodon sample was into two major branches.One of the major ones was mainly composed of the Daxinganling sample and the Xiaoxing'anling sample.There was also a small branch of the Changbaishan Mountains sample(YDFH1,YDHD11,YDHD21,YDHD26)in this large branch.Further suggesting that the Changbaishan Mountains may be the refuge of last glacial for Sorex isodon.The other big branch was mainly composed of samples of the Changbaishan Mountains.The samples from China: Yakeshi and Russia: Sakhalin Island are clustered on this large branch.It is suggested that the Changbaishan Mountains may be the refuge of last glacial for Sorex isodon.(5)The analysis of thr neutrality test and the expected mismatch distributions show that the Sorex isodon population in the Northeast region has no historical population expansion.Research on molecular system geography shows:Get Median-joining network based on Sorex isodon haplotype in Northeast China and GenBank from the GenBank region,the result shows that there were 3 parts.One part was mainly composed of four haplotypes in the Daxinganling and Xiaoxinganling and Changbaishan Mountains in Northeast China(3 yellow from the Hengdaohezi area and 1 purple from the Fenghuangshan area).The other part was entirely from the Changbaishan Mountains in Northeast China.The third part includes the haplotypes of three populations in Northeast China,as well as the haplotypes of Lake Baikal in Russia and the haplotypes in Finland.Three branches all have the haplotypes in Changbaishan Mountains,it is suggested that the Changbaishan Mountains may be the refuge of last glacial for Sorex isodon.
