| Chitosan,the only existing natural cationic polysaccharide prepared from the deacetylation of chitin,has been widely applied in various pharmaceutical sectors including drug release,tissue engineering,oncotherapy and high-efficiency antibiotic with its biocompatibility,biodegradability,hemostasis,antibacterial property,antitumor activity and promotion on tissue recovery.With the chitosan nanoparticles(CNPs)used as active ingredients,not only an improvement in its solubility but also a perfect demonstration of the overview of its outstanding bioactivity and biological pharmaceutical properties could be made.Ionic gelation technique(IGT)is one of the most used ways to prepare CNPs since it can be conducted in a mild reaction condition without organic solvents at room temperature.But the stability of the nano preparations will be influenced by the sendimentation and aggregation of nanoparticles stored for a long time resulted from the inhomogeneity in distribution nanoparticle sizes under this method.A method of ionic gelation technique combined with high-pressure homogenization was introduced into this study to prepare chitosan nanoparticles and investigate their characterization,stability and antibacterial activity.Results of the study were depicted as below:1.In micro-morphological observation,chitosan nanoparticles prepared by ICH method presented a uniform sphere,significantly improving the irregular morphology that caused by ionic gelation technique.2.Benefited from the optimization of parameters of high-pressure homogenization preparation,the average particle size and potential of chitosan nanoparticles prepared by ICH were 180-250nm and 25-35mV respectively which were obviously lower than those of nanoparticles prepared by IGT.It indicated that nanoparticles prepared by ICH could get a lower mean particle size and potential.3.The stability of chitosan nanoparticles prepared by ICH could be further strengthened through optimizing prescriptions of relevant pharmaceutical polymers.When concentrations of P-407,P-188,PEG and HP-β-CD were 1.0 g/L,0.8 g/L,1.0 g/L and 1.0 g/L respectively,no significant change were observed in particle size and potential indicating a good application prospect of the IGT chitosan nanoparticles.4.A Temperature-sensitive gel with controllable temperature and time for gelation and good stability could be prepared with the utilization of chitosan nanoparticles prepared by ICH.5.Chitosan nanoparticles prepared by ICH also showed a good antibacterial activity.A test for their antibacterial activity was conducted with Candida albicans as experimental bacteria and among of which were treated by Miconazole nitrate as positive control groups.Compared with the antibacterial diameter of chitosan nanoparticles prepared by IGT,a higher size could be tested in nanoparticles prepared through ICH.In conclusion,the method of ionic gelation technique combined with high-pressure homogenization set up in this study can help to improve the particle sizes,potential,stability and antibacterial activity of chitosan nanoparticles,demonstrating a better application prospect in preparation products than ionic gelation method. |
