Keratin is the main component of wool,feathers,nail and other epithelial coverings.As a natural bio macromolecule,keratin has good bio activity,biocompatibility,biodegradation and natural abundance.Keratin-based biomaterials have emerged as potential candidates for various biomedical applications.In the study,human hair keratin was extracted to prepare keratin-drug nanoparticles(KDNPs)as drug delivery systems.(1)Preparation of keratin/chlorhexidine complex nanoparticles by ionic gelation technique.These nanoparticles were characterized by UV-Vis spectrum(UV),fourier transform infrared spectroscopy(FTIR),dynamic light scattering(DLS),scanning electron microscope(SEM)and transmission electron microscopy(TEM).Antibacterial test indicated that the nanoparticles had obvious antibacterial activity against staphylococcus aureus.(2)Preparation of keratin/doxorubicin complex nanoparticles by ionic gelation technique.Turbidity test indicated that the stability of the complex nanoparticles was inversely proportional to the amount of drug addition.The doxorubicin release behavior showed that these nanoparticles were both sensitive to pH and glutathione.The results of MTT showed that high concentrations of the complex nanoparticles had obvious anti-cancer properties.(3)Preparation of keratin/doxorubicin complex nanoparticles by desolvation method for adsorption of hydrochloric acid doxorubicin.Keratin nanoparticles were prepared by desolvation method and then adsorbed of doxorubicin hydrochloride.In vitro release experiments showed that the release of doxorubicin was faster in acid and glutathione environment.(4)Preparation of keratin/doxorubicin complex nanoparticles by precipitation method.The keratin and hydrochloric acid doxorubicin was first mixed according to certain proportion,and then keratin/doxorubicin complex nanoparticles were prepared by precipitation method.The experimental results showed that low concentration of nanoparticles had obvious anti-cancer properties. |
