?-mannanase is widely distributed in nature as a substrate hemicellulase,it can be applied in many industrial fields.Bacillus licheniformis HDYM-04 is a high production of ?-mannanase which was isolated in flax retting and demonstrated.The process of enzyme production by HDYM-04 has been further studied in this thesis.The fermentation conditions in batch and fed-batch fermentation were optimized and the effects of carbon source and nitrogen source on production of ?-mannanase in batch fermentation were also studied.Besides,the establishment of the batch fermentation kinetics model lay a solid foundation for the strains used in industrial production.Carbon source optimal conditions of the HDYM-04 fermentation is:initial adding 0.5%glucose and 1%konjac flour;add 1%konjac flour when enzyme activity reduced,add 0.5%when reducing sugar reduced.Nitrogen source optimum conditions of HDYM-04 fermentation is initial adding 0.5%NH4NO3 and 1%yeast flour;adding 0.5%yeast when enzyme activity reduced.This study optimize the fed-batch fermentation conditions of production of?-mannanase by Bacillus licheniformis HDYM-04 in a 5 L fermentor,which lay a foundation for the industrial application of the HDYM-04 and provide a practical guidance.
