Iron Deficiency Anemia?IDA?is caused by various reasons which lead to the lack of iron element stored in body.Iron supplement agents were usually selected to treat IDA,however,iron agents bring about many side effects such as nausea,abdominal pain,diarrhea or constipation,which do more harm to gastrointestinal health,besides their effects of improve anemia are limited.,they are so easy to reappear that there it is needed to seek safe,efficient and low side effects substance for anemia.Our previous studies have found that Auricularia auricula-judae melanin has significant effect on treating the IDA,in order to make the product appear on the market and take benefits to society,its mechanism of treating IDA and its effect on the intestinal health need to be further in-depth study.Melanin was prepared from Auricularia auricula-judae,by the method of acid resolving,alkali dissolving,acid sinking and then purification.The effects of Auricularia auricula-judae melanin on iron deficiency anemia and intestinal micro ecology were deteted through the establishment of IDA mice model,the resuls are as follows:?1?IDA+M group had a significantly higher number of Hb and RBC compared with IDA and IDA+Fe2+group,Hb concentration was 171.67±7.53 g/L,85.00±15.17g/L and 156.67±8.16 g/L respectively;RBC counts were 11.38±1.37 1012/L,8.15±0.741012/L and 8.15±0.92 1012/L respectively;The serm SCF concentration of IDA+M group was 904.01±53.30 pg/mL,serm EPO was 182.28±17.70 pg/mL,serum GATA-1was 13.00±0.70 ng/mL,serum iron was 1.44±0.06 mmol/dL the above index were significantly higher than that of IDA and IDA+Fe2+group?p<0.05?,the above two index of the latter groups were serm SCF 827.31±64.84 pg/mL,64.84±98.13 pg/mL,serm EPO 137.62±11.62 pg/mL,154.65±11.38 pg/mL,serum GATA-1 10.72±1.02ng/mL,10.66±1.62 ng/mL,serum iron 0.75±0.04 mmol/dL,1.09±0.07 mmol/d L respectively,among the detected blood indexes the melanin groups were superior to FeSO4 groups,FeSO4 groups were statistically significant differences?p<0.05?.These indicate that Auricularia auricula-judae melanin could effectively improve the symptoms of IDA,and promote the body's absorption of iron and hematopoietic function.?2?Auricularia auricula-judae melanin could improve IDA mice intestinal microflora abundance falling such as S24-7?norank,Alistipes,Ruminococcaceae?uncultured,Blautia,Ruminococcaceae?unclassified,Prevotellaceae?uncultured,six main species of bacteria,the improve value was 0.01%?3.87%,it could reduce IDA mice intestine,Allobaculum and Parabacteroides abundance of species of bacteria,the reduce values were 21.12%and 3.51%respectively,it could also improve the IDA mice intestinal microflora,Gleason,Pielou and Shannon-wieaver index,better than ferrous sulfate,Control+M group,Control group,Control+Fe2+,IDA+M group,IDA+Fe2+group and IDA group each group Gleason index of intestinal flora in mice,in turn,were3.27,3.07,3.02,3.01,2.88,2.61 respectively,Pielou index,in turn,were 0.578,0.564,0.505,0.502,0.430,0.410 respectively,Shannon-wieaver index,in turn,were 0.866,0.848,0.823,0.827,0.634,0.586 respectively.This showed that Auricularia auricula-judae melanin could make IDA mice intestinal flora changes restoration,reduced the difference between control mice ande IDA mice and get close to normal condition.?3?The beneficial substances short chain fatty acid propionic acid,butyric acid,metabolites in IDA+M mice intestinal were significantly higher than the IDA and IDA+Fe2+group?p<0.05?,the concentration of propionic acid were 5.21±0.96 mg/mL,2.61±0.53 mg/mL,0.53±0.50mg/mL,butyric acid concentrations were 4.27±0.91mg/mL,1.04±0.35 mg/mL,0.35±0.57 mg/mL;And intestinal metabolites of palmitic acid,stearic acid,oleic acid,total amino acids were significantly improved?p<0.05?,palmitic acid concentrations were 1.56±0.17mg/mL,1.22±0.13 mg/mL,1.09±0.12mg/mL;stearic acid concentrations were 1.34±0.15 mg/mL,1.19±0.13 mg/mL,0.92±0.10 mg/mL;oleic acid concentration were 0.46±0.04 mg/mL,0.23±0.03 mg/mL,0.34±0.04 mg/mL;Total amino acids were 23.91±2.31 mg/mL,17.00±3.42 mg/mL,22.56±1.71 mg/mL.These showed that Auricularia auricula-judae melanin could improve the concentration of IDA mice intestinal metabolites,decrease the difference between the IDA mice and the control mice,make the intestinal metabolites tend to be normal.
