Objectives To prepare the RES-BSANP,and to explore the effect and mechanism of RES-BSANP on the proliferation and apoptosis of human hepatica cell line HepG2 in vitro.To provide new ways for RES' clinical application and cancer clinical care.Methods(1)Desolation the Chemical cross-linking was used to prepare RES-BSANP,also albumin encapsulation efficiency and drug loading effici ency were determined by high performance liquid chromatography(HPLC).(2)First,in order to observe the effect of RES-BSANP on HepG2 proliferation,positive control 5-Fu group(final concentrations are 10 ? 20 ? 40 ? 80 ?g/mL),RES group(final concentrations are 10?20?40?80 ?g/mL),RES-BSANP GROUP(final concentrations are 10?20?40?80 ?g/mL),blank albumin nanoparticles group and a blank control group were set.CCK-8 detection method was used to detect THE effect of RES-BSANP on HepG2 proliferation.(3)To observe the effect and mechanism research of RES-BSANP,which leads to HepG2 cell lines apoptosis,positive control 5-Fu group(final concentration is 40?g/mL),RES group(final concentration is 40?g/mL),RES-BSANP group(final concentration is 40?g/mL),blank albumin nanoparticles group and a blank control group were set.By using RT-PCR,the effect of RES-BSANP to lead to HepG2 cell lines apoptosis and the influence the expression level to related gene Caspase-3,back-2,Bax was observed.For the data comparison one-way analysis of variance and LSD was used.Data has a statistical significant difference if P<0.05.Results.(1)Using desolation the Chemical cross-linking to prepare RES-BSANP,the average drug loading efficiency is 5.97% and average en capsulation efficiency is 37.4%.(2)Compared to the blank control group and blank nanoparticles group,both RES-BSANP and RES group show suppression on the HepG2 proliferation,and the suppression increases by the concentration.The difference has statist ic meaning(p<0.05).Compared to positive control 5-Fu group,RES has the almost same results for HepG2 cell lines apoptosisand the difference has no statistics meaning(p>0.05).The apoptosis of RES-BSANP results is lower than positive control group,and the difference has statistics meaning(p<0.05).Compared to RES-BSANP and RES group,RES-BSANP results is better than RES group which has statistics meaning(p<0.05).(3)Compared to blank control group and blank nanoparticles group,positive control 5-Fu group,RES-BSANP and RES are up-regulation the HepG2 cell lines that boost apoptosis gene Caspase-3,Bax,Bcl2 mRNA.The results are almost the same,which has no statistics meaning(p>0.05).The results are statistically significant lower by RES(p<0.05).Conclusion The RES-BSANP preparation technics is reliable.RES-BSANP has statistically significant impact on theapoptosis of HepG2 cell lines proliferation,and the apoptosis function is the same as 5-Fu.This function mechanic may be related by the Caspase-3,Bax,Bcl2 mRNA gene expression.
