Drug Molecule Cyclodextrin Inclusion Phenomenon And Nano-particle Construction And Purification Process Of Azadirachtin

The thesis is mainly divided into three parts,respectively,characterization and in vitro evaluation of the two morin/CD inclusion complexes,characterization of the new targeted oxaliplatin delivery systems based on synthesized two ?-CD modified hyaluronic acid,study on the purification process of neem by "flash column method".In the first chapter,we studied the inclusion behavior and biological activity of natural morin with two kinds of cyclodextrins inclusion complexes.In a review of cyclodextrin as commonly small molecular of drugs inclusion carrier on the basic research of mechanism and behavior,we successfully prepared this kind of insoluble natural morin with two cyclodextrins(amino-P-CD and ethylenediamine-?-CD)inclusion complexes,and systematically investigated in both aqueous solution and the solid state by means of UV-Vis spectroscopy,1H NMR,2D NMR(ROESY),SEM,FT-IR,DSC,TGA,XRD analysis and Molecular Modelling.The MH/cyclodextrin complexes still maintained antibacterial activity in vitro test against Staphylococcus aureus bacteria.In the second chapter,following the inclusion behavior of cyclodextrin with small molecule drugs and combined with our previous study on the inclusion behavior of cyclodextrin with oxaliplatin,we choose to use hyaluronic acid and cyclodextrin(ethylenediamine-?-CD and hexamethylendiamine-?-CD)by amide condensation reaction to construct the amide bond,then,prepare the inclusion of nanoparticles with oxaliplatin and apply the UV-Vis spectroscopy in the phenolphthalein method,NMR,TEM,SEM,AFM,AAS,enzymatic reactions and DLS to verify the grafting ratio,inclusion pattern and enzymatic release of HACD-OXA.The third chapter is mainly to carry out the research on the purification technology of azadirachtin by "flash column method".In the review of the background and the domestic and foreign research of development of azadirachtin in the extraction and purification process,we use the first crushing shell,after the first leaching extraction degreasing,azadirachtin directly precipitated yellow-white powder,then,purified by"flash column method" and screened azadirachtin optimal elution system and stationary phase by HPLC,provided the further development of neem industrial mass production basis quickly.
