Cervical cancer is one of the most common gynecological malignancies,and its incidence is extremely high in developing countries.Compared with synthetic compounds,natural compounds are purely natural and have less toxic and side effects when treating tumors.Portulaca oleracea.L is a weed widely distributed at the edge of fields and vuins.Previous studys have showed that Portulaca oleracea.L polysaccharide has a strong anti-cervical cancer activity and its mechanism of action is clarified.As a functional drug with green environmental protection type,Portulaca oleracea.L has great potential for its development and utilization.Decoction is a common oral dosage form of Chinese herbal medicine,but the decoction has disadvantages such as boiling,carrying and storage.However the granules have the advantages of fast absorption,portability and high biostability.In this experiment,ultrasonic extraction was used to extract polysaccharides from Portulaca oleracea L.(POL-P).The effects of extraction time,extraction temperature and ratio of material to liquid on extraction rate were studied by single factor experiment,and orthogonal experiment was designed according to single factor experimental results.20 Kunming mice(male and female)were used for acute oral toxicity test.Morphological changes of liver and kidney were observed by HE staining.The fuctional changes of liver and kidney in mice were detected before and after acute toxicity test.The mice model of cervical cancer were established,and the experiment was divided into four groups including POL-P low dose group(POL-P(L)),POL-P high dose group(POL-P(H)),negative model group and positive control.After 13 days of continuous administration,the tumor block was taken to calculate the tumor inhibition rate.The effect of POL-P on the proliferation activity of Hela cells was detected by MTT method.In order to control the quality and stability of POL-P granules,preliminary screening of single excipients was carried out though moisture absorption rate,particle size pass rate and the angle of repose.The suitable wetting agent concentration was screened by soft material,molding rate,angle of repose and granule state after drying.The optimal preparation process was selected by taking the dosage ratio of pharmaceutical powder and auxiliary materials,the ratio of different auxiliary materials and the concentration of wetting agent as the investigation factors.The comprehensive evaluation included hygroscopicity,molding rate and angle of repose.The results showed that the best conditions for POL-P by ultrasonic extraction was 55 min for extraction time,45? for extraction temperature,and 1:35 for ratio of material to liquid.Acute toxicity test did not cause acute death or other abnormalities in mice within one week.Liver and kidney were not damaged and blood was normal.POL-P has a significant inhibitory effect on solid tumors.POL-P inhibited Hela cell proliferation in a dose-and time-dependent manner.The results of preparation and evaluation criteria for POL-P granules showed that power(1:1.5),lactose and soluble starch mixed adjuvant(2:1),using 80% ethanol as humectant.According to the four general rules 0104 with granules in the 2015 "Chinese Pharmacopoeia",the property,grain size,moistune and solubility of POL-P granules were all conform with the standard.This study provides theoretical basis for the development of POL-P as a new anti-cervical cancer drugs.It is also the technical guidance for the industrial production of POL-P granules.
