This thesis mainly from the multifunctional nano MOF material preparation,application and the detection of tumor markers,using fluorescence,uv absorption and laser scanning confocal microscope technology,based on the synthesis of metal organic framework material material,through the functional resorcinarene,the assembly is applied to the treatment of tumor cells,and in quantitative analysis.At the same time,ATP detection and in vivo experiments on mice were carried out.The main contents of this paper are as follows:1.Application of MOF in the identification of active molecules.Three-dimensional lanthanide MOF?Ce-MOF?was constructed from amino functionalized ligands and Ce³⁺ions by one-step synthesis.ATP was detected at the nanometer level by the construction of ATP-Ce-MOF.This is an innovation of electrochemical sensor based on MOF to detect ATP.This method has been successfully applied to the detection of ATP in serum of tumor patients,and has potential application value in clinical diagnosis.2.Application of MOF in drug loading.We propose a variety of DNA and FA double-functionalized nano-mof composite materials that can achieve targeted tracking and targeted treatment of tumor cells.A pH responsive controlled release system for combined drug delivery and targeted therapy was constructed.In UMCM-1-NH₂-FA-DNA₂-DOX,FAM fluorescently labeled DNA was bound,folate target recognition elements and drug molecules were overexpressed in some tumor cells,DNA functionalization was modified with fluorescein,UMCM-1-NH₂ was used to track tumor cells and cell imaging,and pH response was used to control drug release.Modification of folic acid?FA?results in targeted selectivity of functional materials.3.Metal-organic framework for DNA functionalization:tumor cell tracing,targeted drug delivery and pHotodynamic therapy.We proposed a DNA functionalized nano-metal-organic framework material that can transport DOX and Metal-Organic Framework containing pHotosensitizer to tumor cells via drug delivery and pHotodynamic therapy to target A549 cells.The results showed that the combined drug delivery platform?chemotherapy?and pHotodynamic therapy?PDT?were more effective than the single drug delivery platform.In addition,PCN-224-DNA-DOX can also be used as a probe for the detection of tumor cells.To sum up,PCN-224-DNA-DOX based co-administration,pHotodynamic therapy,targeted platform and diagnosis-therapy integrated system will also become a more promising tumor treatment.DNA functionalized NANO-MOFs can track tumor cells and target drug delivery and Photodynamic therapy.The strategy also showed good performance in the qualitative and quantitative analysis of specific ligands of intracellular nucleic acids and aptamers.
