In post-translational modification(PTM),histone proteins acetylation by histone acetyltransferases(HATs)is an essential regulatory mechanism in epigenetic mark system,which controls the structural organization and transcriptional status of chromatin.Furthermore,it also plays an important role in many fundamental biological processes,such as DNA replication and repair,genome stability,histone deposition and nutrient metabolism.Many studies showed that aberrant HATs activities have been associated with the pathogenesis of many diseases,such as cancers,neurological disorders,HIV infection and inflammatory,etc.,and HATs have emerged as significant biomarkers for these diseases,and its inhibitors contribute to the chemotherapy as well as drug discovery of anti-cancer agents.As a result,reliable approaches for detecting activity of HATs and screening of its inhibitors are of paramount importance for clinical diagnostics,biological medicine and biochemical research.Electrogenerated chemiluminescence(ECL)biosensors have unparalleled advantages such as high sensitivity,simple equipment and easily operation,fast detection,easily control and low cost.In recent years,scientists have come to an endless stream of research by using ECL biosensor.By the development of various signal amplification approaches and signal conversion mechanism,we studied the combination of signal amplification approach and ECL luminophor,and the change of ECL reaction induced by another reaction.Finally,they are used for HATs activity detection and its inhibitors screening.An ECL signal probe was prepared by supersandwich DNA stabilized silver clusters,and the supersandwich DNA was formed by the hybridization chain reaction(HCR)of DNA.Finally,the probe was used for the HATs activity detection.From the results,the biosensor showed fast response and high sensitivity.A precise detection also can be carried out after inhibitor stimulation and the biosensor can be used for inhibitor screening of HATs.Even in the cell lysate sample,the detection of HATs activity also can conduct by this biosensor.A sensitive turn-off ECL biosensor based on the superoxide dismutase(SOD)performance of tannic acid(TA)was developed.In this strategy,TA can eliminate superoxide radical,which is co-reactant of luminol ECL,and induce reduced ECL signal as the reflection of HATs activity.The biosensor also can be used for HATs activity detection in cell lysate successfully.
