Purpose:1.The preparation of the plantasanOptimize the best process to extract the medicine of plantasan,a series of methods of removing impurities were used to obtain the purified plantasan.2.Quality standard of the plantasanEstablishing a pre-column-derived HPLC method and UVspectrophotometry to determine the content of polysaccharides and monosaccharide composition.Applying this method to systems adaptation experiments to examine its precision,stability,etc.3.Experimental reseacrh of plantasan on immunological function in immunocompromised mice To examine the effect of plantasan on immunological function in immunocompromised mice.Methods:1.The preparation of the plantasanWater extraction ethanol precipitation process is adopted to extract the plantasan,proteins were removed by Sevage method,decolorize by hydrogen peroxide,finally,the plantasan was purified by washing with organic solvent such as acetone.2.Quality standard of the plantasanThe samples were treated by PMP column derivation and hydrolysis,etc.Using Agilent C18 HPLC column?4.6 mm×250 mm,5?m?,The flow rate was 1.0 mL·min^-1,the detection wavelength was 245 nm,the moving phase was phosphate buffer(0.1 mol·L^-1)and acetonitrile,HPLC was used to analyze monosaccharide composition and determine its content.3.Experimental reseacrh of plantasan on immunological function in immunocompromised miceImmunosuppressive model in mice was established by abdominal injeccting cyclophosphamide,A large,medium and low dose of plantasan (900/300/100mg·kg^-1)was administered by gavage for 21 days,The effects of immune function was observed by detecting the corrected carbon particle clearance index,poor thickness of toe,weight gain rate,spleen index,thymus index and half hemolytic value in mice.Results:1.After 12,10,8 times water extraction 3 times,add an equal amount of ethanol to soak it twice,and then the crude extract of the soil-gray fibrous plantasan wasobtained,after refinement,the plantasan obtained is white fine powder.2.The hydrolysis product of plantasan is a kind of acid polysaccharide composed of mannose,rhamnose,glucuronic acid,galactose,glucose,galactose,xylose andarabinose.The content of its monosaccharide component was between 1.636⁹1.19mg·g^-1,and the result of methodology verification is good,It all fits the criteria.3.Plantasan could significantly imporve the delayed hypersensitivity in mice,inerease the hemolysin levels in immunocompromised mice,enhanced the carbon particle clearance function in mice,and increased the weight,spleen index and thymus index.Conclusion:After the best process extraction of plantasan,the yield was 32.21%.Based on the analysis of monosaccharide composition,total sugar content and water content,the preliminary quality standards were established.Through the experiment of correcting carbon particle clearance index and other indicators in mice,it is proved that plantain glue has certain function of strengthening immunity.
