At present,there are many new methods of cancer treatment.Recently,the new treatment technologies are new drug chemotherapy,PDT,PTT,MHT,embolization and gene therapy.However,due to the lack of receptor density on the surface of cancer cells,which limits the ability of tumor cells to actively ingest nanoparticles,the treatment effect is not good and the side effects are serious.In order to solve this problem,a promising self-amplified apoptosis targeting strategy is proposed,which is to generate more targets in the dynamic process of targeting and treatment,so that our nano therapeutic agents can target the focus more efficiently.Compared with the traditional photothermal therapy,magnetic thermotherapy has attracted more and more attention because it is non-invasive or minimally invasive and does not have tissue penetration.Magnetic hyperthermia uses magnetic thermal effect to kill pathological tissue cells with high temperature.That is to say,the magnetothermal material in alternating magnetic field produces a lot of heat under the effect of magnetic field radiation,which can rapidly increase the temperature near the material and kill the pathological cells.The characteristics of accurately locating and killing the diseased cells or tissues make magnetohyperthermia stand out in many treatment technologies,and more and more attention has been paid.At the same time,the new drugs are delivered to the diseased tissue by carrier,and released slowly in the environment of tumor micro acid,which can reduce the dosage of drugs,thus greatly reducing the toxicity and side effects of the whole body.The combination of magnetic hyperthermia and chemotherapy can further improve the effect of cancer treatment.Compared with the first mock exam,the multimodal treatment has the advantages of single mode and even synergy,and its therapeutic effect is greatly improved.Nano-materials have a series of unique acoustic,optical,magnetic and thermal properties.In vivo imaging,they can be used to evaluate tissue function or disease diagnosis,and can be used in US,PAI,MRI,pet,SPECT,CT and other medical imaging.Magnetic resonance imaging?MRI?is widely used in soft tissue imaging because of its high spatial resolution and penetration ability.The use of MRI contrast agent is convenient to distinguish tumor tissue and normal tissue in the process of tumor treatment,which provides a very useful basis for the study of tumor treatment.Inspired by the apoptotic targeting properties of phosphatidylserine?PS?,combined with the excellent magnetic properties of Zn0.4Co0.6Fe2O4@Mn0.4Co0.6Fe2O4 nanoparticles?MNPs?,a self-amplified targeted apoptotic nano-platform?MNPs-ZnDPA/?-Lap?was prepared.?-Lapachone??-Lap?is a chemotherapeutic drug,which can promote apoptosis.Zn?II?-bis?dipicolylamine??bis-ZnDPA?is a good targeting part of apoptosis,and magnetic field radiation is used as external energy.In the apoptosis 4T1 xenotransplantation model,MNPs-ZnDPA/?-Lap first accumulated in the tumor through EPR effect.The?-Lap released after reaching the tumor triggered the apoptosis of tumor cells and increased the apoptosis target,which enhanced the apoptosis target characteristics of nanoparticles.On the other hand,the temperature of MNPs increased under the condition of external magnetic field to kill tumor cells.This self-amplified apoptosis targeting effect of MNPs-ZnDPA/?-Lap can almost inhibit the growth of tumor through coordinated magnetothermal/chemical therapy,which provides an important prospect for targeted cancer therapeutics.
