Methotrexate?MTX?is a folic acid antimetabolite,has strong immunosuppressive effect,is one of the clinical basic anti-tumor drugs,has become used in leukemia,lymphoma and other malignant tumors and psoriasis and other autoimmunity A commonly used antimetabolite for disease.However,the toxic and side effects of MTX are relatively large,mainly reflected in bone marrow suppression,mucosal reaction and liver and kidney damage.This defect has seriously affected the application of drugs.How to better improve the curative effect of drugs and reduce their side effects has become a problem that researchers have been exploring.Recent studies have shown that the original anti-cancer drugs are loaded on a suitable drug carrier to form a new anti-cancer drug carrier composite material,so as to improve the bioavailability of the original anti-cancer drugs and reduce physiological toxicity.Purpose is a very effective way.Among various types of drug carriers,inorganic nano drug-carrying systems have become the focus of research due to their many preparation methods,easy morphology control,good biocompatibility,stable performance,and high loading capacity.In this paper,the commonly used anti-tumor drug methotrexate?MTX?is used as the guest drug,and three types of MTX/inorganic nanocarrier complexes are systematically prepared,and their structures,properties,sustained-release properties of the drug,and the Their inhibitory effect on Hela cells.The main research includes:1.Modified attapulgite material?ATP?,compounded with precious metal nanomaterials to obtain ATPAu nanorods,and compounded with easily degradable polymer nanomaterials,successfully prepared ATP?H+?with good stability and excellent slow release performance/MTX drug loading system.Through TEM,XRD,UVVis,FTIR,Zeta and other characterization methods,a good compound between the drug and the carrier is proved.The MTT cell experiment proved that ATP has no biotoxicity to Hela cells,and the ATP?H+?/MTX complex has a good inhibitory effect on Hela cells.The effect of light on the anticancer effect of ATPAu nanorods was further investigated through experiments.The results showed that the inhibitory effect of ATPAu nanorods on Hela cells was attributed to the photothermal effect of gold nanorods.Studies have shown that ATPAu nanorods are an effective drug carrier,which can be further studied and explored in the future on the effects of light and heat.2.Select MTX as the guest drug molecule and Au@mSiO2as the drug carrier to realize the loading of MTX on Au@mSiO2by electrostatic attraction,and prepare the Au@mSiO2/MTX drug-loading system.Gold nanoparticles were synthesized by seed growth method,and the silica layer was coated by adding Na OH and TEOS.After adding absolute ethanol and concentrated hydrochloric acid to remove CTAB,the mesoporous silica layer is coated on the surface,and MTX is loaded on Au@mSiO2by electrostatic attraction,thereby obtaining the drug loading system Au@mSiO2/MTX.A series of characterization methods such as TEM,XRD,UVVis,FTIR,and Zeta prove that the prepared Au@mSiO2/MTX composite has regular morphology and good dispersion.The drug loading of Au@mSiO2and the sustained release performance under different p H conditions were further studied.Through a comparative study of the cytotoxicity of MTX,Au@mSiO2,Au@mSiO2/MTX,Au@mSiO2/CTAB,it was found that Au@mSiO2CTAB is the most toxic,so it is very important to remove residual CTAB during the preparation process.Studies have shown that the Au@mSiO2/MTX system has a good inhibitory effect on Hela cells,and at the same time Au@mSiO2does not have too many toxic and side effects on Hela cells.The biological experiment provides basic data.3.Using MTX as the guest drug molecule and hydroxyapatite?Hap?as the drug carrier,HAp/MTX complex was prepared by in-situ method,and the characterization results showed that the complex was successfully prepared.Due to the impact of the epidemic,the subsequent performance experiments of the HAp/MTX complex could not be carried out systematically.This article prepared three types of MTX/inorganic nanocarrier complexes of ATP?H+?/MTX,Au@mSiO2/MTX,HAp/MTX through different methods,and discussed in detail the structure and properties of the first two systems and their inhibition on Hela cells Effect,the results show that MTX/inorganic nanocarrier complex has a good inhibitory effect on Hela cells,while the toxic and side effects are significantly reduced,and it has a good application prospect.
