Amorphophallus konjac is the perennial herbaceous crops of Amorphophallus Blume and cultivated extensively in the southern part of China.Konjac contains rich glucan-mannan which has various biological activities and extensive application value.Konjac had cultivated widely in many regions of Guizhou province with the inereasing demand for konjac products.And the government is working on creating konjac industry chain in Guizhou.However,konjac soft rot has devastating effect on konjac planting that restricts severely the development of konjac industry.Mang researchs about prevention and treatment of konjac sof rot had done and having a certain effect,but the problem is not solved fundamentally.And the pathogens of konjac soft rot has not been researched in Guizhou province.In this work,the isolation,identification,infection characteristics,the technical aspects of prevention and treatment of soft-rot konjac from Taijiang County of Guizhou were discussed.The main results were as follows:1)The pathogenic strains of Amorphophallus konjac from Taijiang County,Guizhou were isolated,inoculated and identificated.One pathogenic bacteria strain to cause Amorphophallus konjac soft rot is isolated from 16 strains.The pathogen was identified as Erwinia chrysanthemi by 16 S rDNA sequence analysis.2)The engineering strain with GFP marker is successfully obtained by chemical conversion method.3)Getting tissue culture konjac plantlets through tissue culture.4)Pathogen infection characteristic findings show that the pathogen coundn’t infect the konjac through natural orifice and roots,mainly through the wound to infect konjac,and via the veins,petioles vascular to diffuse.The konjac fleshy petioles and mesophyll were infected mainly,the epidermis is not easy to be destroyed.And a small amount of pathogens can not cause the soft rot,only the pathoges reaches a certain number.5)The incidence of konjac soft rot and konjac yield without certain relations of increasing and decreasing with the biochar were found through potted plants and field experiments.
