Fragrance is one of the most significant rice qualities.Improvement of rice fragrance quality will undoubtedly enlarge rice marketing value.Rice fragrance was suggested to be controlled by a betaine aldehyde dehydrogenase(OsBadh2)gene,which is located in rice chromosome 8.The disfunction of this gene will lead to massive production of an aromatic compound 2-acetyl-1-pyrroline(2AP)in rice,which consequently generate rice fragrance phenotype.This study applied a recently developed gene-editing technology,which is mediated by a transcription activator-like effector nuclease(TALEN),in result,capable of modifying the target gene.Through TALEN technology,we successfully disenable the OsBadh2 gene in an elite japonica variety H506,strongly adding the material the rice fragrance.The main results were concluded as follows.1 A functional TALEN vectors was established.2 Through transformation of TALEN via agrobacterium mediated genetic transformation,we obtained 1000 independent hygromacin-resistant calli,and among them,732 independent transgenic plants were differentiated.3 Through analysis by sequencing the TALEN aimed site in the target gene OsBadh2,we identified 2 plants,in which several bases were occurred deletion,leading to gene frame shift mutation.The homologous lines were able to produce readily perceivable fragrance.
