Gladiolus hybridus is a perennial bulbous ornamental plant of the Iridaceae Gladiolus,and the Gladiolus cormel takes three years to develop a flowering ball.Gladiolus cormel has dormancy characteristics,which can be broken through certain processing in the facility environment.By shortening time of the cormel turning to the ball,the economic benefits can be increased.In order to provide practical process with reference,we need to study the effective lifting of the cormel dormancy method to promote germination.In this study,three kinds of gladiolus cormel varieties 'Super Red','Pink Diamond','White heaven,were used as test materials.In order to test the changes of physiological indicators during the breaking-dormant process,we used different concentrations of pharmacy to treat the cormels in four different stages of dormancy.The main conclusions are as follows:(1)Different gladiolus cormel has different dormancy type.All the species begin its dormancy duration at the 60th day after being harvested,while the time of releasing dormancy is different.In addition,the 'Pink Diamond' and 'Super Red'which belong to the type of moderate physiological dormancy can release its dormancy at the 90th day after being harvested;However,the specie of 'White heaven' can't release its dormancy after 150 days which means this kind of gladiolus cormel belongs to a type of deep physiological dormancy.(2)The effect of break dormant treatment on cormel dormancy was different.By treating 'White heaven' with 1 mg/L TDZ after being harvested,we found that the germination percentage of this kind cormel improved 68.88%than the control treatment.Another treatment which is the best method and can totally break the cormel dormancy was using 100 mg/L GA3 to treat 'White heaven' at the 90th day after being harvested.However,both treatment have no influence on 'Super Red' and'Pink Diamond'.Furthermore,after entering the dormancy termination period,all the treatments were inhibited to varying degrees except 100 mg/L GA3.(3)From the effects of pharmacy on the sprouting effect of three varieties of gladiolus cormel,there were differences among the agents,and the overall effect was from GA3>TDZ>6-BA>KNO3.Three species cormel's best drug treatment time is as follows:Treating 'Super Red' with 50 mg/L GA3 and treating 'Pink Diamond' with 150 mg/L GA3 at the 60th day after being harvested can both improve the germination percentageten to 50%in ten days' time;While compared with control treatment,treating 'White heaven' with 100 mg/L GA3 at the 90th day after being harvested can shorten the breaking-dormant process at least 60days.(4)Soluble sugar,total amylase activity,?-amylase activity,the soluble protein content and saturated water content,malondialdehyde content,relative conductivity,antioxidant system POD activity,SOD activity and other related substances were related to the cormel germination percentage.In addition,the starch content and PPO enzyme were related to the germination percentage.Compared with the contrast material,the germination rate increased,accompanied by the decrease of starch content and the increase of PPO activity and CAT activity.The soluble protein content was an important factor affecting the germination of cormel.
