Fusarium oxysporum is a type of facultative parasitic fungus that can invade plants and live in the soil.This type of fungus also has subspecies differentiation and has various specialized form.Fusarium oxysporum can parasitize a wide range of plants to induce progression of blight,including more than 100 types of plants like Solanaceae,Leguminosae,melons and so on.Fusarium oxysporum f.sp.Lycopersici(FOL)FGSC 9935 causes vascular wilt disease resulting in huge economic loss for global tomato production.Currently,studies related to tomato wilt disease are mainly focused on cultivation of resistant variety.However,there are a few of researches devoting to explore molecular mechanisms related to resistance to Fusarium oxysporum.1 FRG3,a target of slmiR482e-3p,provides resistance against the fungal pathogen Fusarium oxysporum in tomato.Nucleotide-binding site-Leucine-reach repeats(NBS-LRR)proteins are R proteins encoded by plant resistance genes(R),with largest among R protein families in the plant.This research focused on the function of resistance gene FRG3 during the process of resistance to tomato wilt disease.(1)FOL was used to infect the resistant Motelle(Mot,12/12)and susceptible Moneymaker(MM,i2/i2)with near-isogenic cultivars.Four libraries including MM_H2O,MM_FOL,Mot_H2O and Mot_FOL were generated in the study.Combined with analysis of sRNA-seq data by using bioinformatics an Northern blot,results showed that expression of slmiR482 family was regulated by FOL.(2)Taken together,analyses of sRNA Northern Blot and qRT-PCR exhibited that slmiR482-3p was repressed in Motelle.(3)Bioinformatics analysis indicated that FRG3 was a target of slmiR482-3p and encoded a NBS-type resistance protein containing coiled-coil(CC)and P-loop domains.(4)Phylogenetic analysis showed that FRG3 was not a homolog of the 12 family,leading us to propose that FRG3 acted as a disease resistance partner to compensate the potential cost of 12 homolog expression to fitness in tomato.(5)Implemented Agrobacterium-mediated transient co-expression experiments in N.benthamiana,it was confirmed that FRG3 was regulated by slmiR482e-3p at the transcriptional level.(6)The majority of slmiR482e-3p cleavage of FRG3 occurred after tenth nucleotide at the end of 3’.(7)The virus induced gene silencing(VIGS)method was used to partially silence FRG3 in the resistant cultivar Mot.Results showed that the decrease in FRG3 expression could result in attenuating the resistance of Motelle to FOL,which indicated that FRG3 played an important role in the resistance to tomato wilt disease.In conclusion,this study revealed a new gene FRG3 was regulated by slmiR48e2-3p at the transcriptional level,which participated in the resistance to tomato wilt disease.This research provided molecular mechanisms for exploitation of resistance to tomato wilt.2 The developing of a rapid detection of the biomass of Fusarium oxysporum in tomatoTomato wilt disease is a type of worldwide soil borne fungal diseases.Fusrium oxysporum can survive in soil during winter.Once infecting the host plant,the pathogen can spread from the bottom to the top in the vascular bundles of plants.Currently,the main prevention and control method is to select the resistant cultivar,but there is no highly resistant variety so far.Therefore,it is important to predict the tomato wilt disease.This study focused on exploring the relationship between the progression of tomato wilt disease and pathogen accumulation in tomato using the susceptible cultivar Moneymaker(MM).(1)FOL was used to infect MM,which made plant growth and vascular bundles affected by pathogens.(2)A standard curve of FOL biomass was establish by using qPCR to evaluate the biomass of pathogen in tomato.(3)The biomass of FOL in the plant was quickly and accurately detected by qPCR.Thus,the relationship between the biomass of pathogens and occurrence of tomato wilt disease was established.This study provided some practical value for guiding prediction of tomato wilt disease.
