Tobacco brown spot resulted from alternaria fungi disease is a devastating disease for tobacco leaves.At present,both at home and abroad the research to Alternaria fungi pathogenic ability and the relationship between signal transduction system mainly concentrated in the two-component signal transduction system(TCSTS)and mitogen-activated protein kinase(MAPK)signal transduction system.In this paper,the related genes of two types of signal transduction system have been studied:(1)In high osmolarity glycerol(HOG)pathways,HOG1 kinase is one of the MAPK,at the same time in the fungal cells ability playing an important role to cope with pressure and stimulating.Its corresponding gene deletion strains was obtained by the method of homologous recombination and completed a series of tests on the research.The results revealed that the AlHOG1 involved in multiple physiological process of Alternaria longipes,including hypha and conidium growth,multi-factor sensitivity,melanin production and nutrition.(2)In the past study,the third class histidine kinase(AlHK1)belonged to TCSTS has been proved to regualted the pathogenic ability of A.longipes.In order to illustrate its pathogenic ability and the role of AlHK1 in the pathogenic process,the time-course was carried out through the extraction of RNA and the RNA-seq was assembled 22345 unigens to analyze.Most shared differentially expressed genes(DEGs)expression pattern,gene ontology(GO)abundance and the number of DEGs with the rising trend in the cluster were analyzed.Finally,the data suggestes that the changes of C-00 infection mainly existed in the early and middle infection,as the changes of gene expression of HKA4 focused on the late infection stage.(3)Transcriptome result shows that a aspartic acid protease(AlPep1)may be regulated by AIHK1 and participated in the tobacco diseases pathogenic process.Using the method of homologous recombination,the corresponding gene deletion strains was obtained and the results contrasted with the wild type strain have showed that AlPepl is a novel pathogenic factor.In addition,AlPep1 still mediated multiple physiological processes of A.longipe,including mycelial growth and spore production,a variety of stress response sensitivity,pathogenicity and nutrition,etc.
