With the increasing density of sea cucumber culture,water quality management has become an important factor restricting the healthy cultivation of sea cucumber.The team developed a new type of water quality control equipment for the problems in sea cucumber ponds—The Nourishing Water Machine System.In this experiment,the effects of three time allocation methods on the regulation of sea cucumber culture ponds were analyzed and compared.They were: the pond running for 18 hours,the pond running for 12 hours and the pond running for 6 hours.In this experiment,water temperature,salinity,dissolved oxygen,microbial diversity and bacterial community structure in water and sediment were selected as monitoring indicators to monitor the three allocation modes of sea cucumber culture ponds annually.The following results are obtained:(1)Variation range of water temperature in three kinds of ponds:2.15 ~ 32.40?,Range of salinity change:24.52 ~ 35.48‰,Range of dissolved oxygen:4.85 ~ 10.85mg/L.There was no difference in water temperature and salinity between the three ponds,but the difference of dissolved oxygen was not significant.The annual average concentration was 18 h,12 h and 6 h in turn.(2)The results of microbial diversity and bacterial community structure in three kinds of pond water are as follows:Bacterial abundance,diversity and homogeneity were the best in 12-hour running pond water;The change trend of bacterial phylum in three kinds of pond water is the same,The variation ranges of ginseng pool for 6h,12 h and 18 h operation are 22-27 gates,19-28 gates and 21-34 gates,respectively,The bacteria in three kinds of pond water are mainly composed of 12 categories,which are Proteobacteria?Bacteroidetes?Cyanobacteria ? Actinocteria ? Verrucomicrobia ? Firmicutes ? Planctomycetes ? Acidobacteria ?Gemmatimonadetes?Latescibacteria?Fusobacteria?Nitrospirae,In addition,the content of 21 phyla bacteria in 54 samples was less than 1%.The first four phyla were dominant,accounting for 95%-99% of the total bacteria.The three ginseng pools were Proteus,the first dominant phylum except Dec.30.The annual abundance of Proteus varied from 24.84% to 52.52%.The annual average values were 18 h,12 h and 6 h,Bacteroides was the second dominant phylum,and the annual abundance varied from 9.39% to 44.62%.The annual average values were 18 h,52.52% respectively.At 12 h and 6h,Cyanobacteria was the third dominant phylum,with annual abundance changing from 7.54% to 60.42%.The annual average value was 6h,18 h and 12 h,and actinomycetes was the fourth dominant phylum,with annual abundance changing from 1.45% to 27.01%.The annual average value was 6h,12 h and 18 h,respectively.Pearson correlation analysis showed that Bacteroides were negatively correlated with temperature in three kinds of ginseng pools,Cyanobacteria was positively correlated with temperature in 18 h and 12 h ginseng pools,Proteus was positively correlated with dissolved oxygen in 18 h ginseng pools,nitrate was positively correlated in 6 h ginseng pools,and Actinomycetes were positively correlated with organic matter in all three kinds of ginseng pools.The number of bacteria in the water of the water heater had the same trend.The variation ranges of the ponds were 192-335 genera,169-345 genera and 141-351 genera for 6h,12 h and 18 h running of the water heater respectively.PCA analysis showed that the bacteria in the water environment began to play a role after 12 h running of the water heater.(3)The results of microbial diversity and bacterial community structure in the sediments of the three kinds of water-raising machines are as follows: the bacterial abundance,diversity and homogeneity in the water of the water-raising machines running for 12 hours are the best in most of the time,and the change trend of bacterial phylum in the sediments of the three kinds of water-raising machines is the same.The variation ranges of ginseng pools were 35-46,36-45 and 35-47 after 6h,12 h and 18 h operation respectively.The bacteria in the water of the three ginseng pools were mainly composed of 13 categories,namely Proteus,Bacteroides,Chloroflexi,Actinomycetes,Verruca,Phytophthora,Phytophthora,Cyanobacteria,Acidobacteria,Bacillus,Latescibacteria,Clostridium,Nitrospiruli.Mushroom door.The other 37 species of bacteria were less than 1% in 54 samples.The first four bacteria are dominant,accounting for 69.4%~86.6% of the total bacteria.Among the three species,Proteus was the first dominant phylum with an annual abundance change of 43.72%-63.22%.The annual average value was 12 h,18 h and 6 h.Bacteroides was the second dominant phylum with annual abundance change of 6.12%-18.01%.The annual average value was 18 h,6 h and 12 h.Curvularia was the third dominant phylum with annual abundance change of 3.17%-23.64%.Actinomycetes was the fourth dominant phylum with annual abundance change of 3.17%-23.64%,6 h,18 h and 12 h.The change was 1.19%~8.79%,and the annual average value was 18 h,12h and 6h respectively.Pearson correlation analysis showed that Proteus had a positive correlation with temperature in the 6h ginseng pool,Bacteroides had a positive correlation with temperature in the three ginseng pools,Curvularia had a positive correlation with temperature and dissolved oxygen in the three ginseng pools,Actinomycetes had a significant positive correlation with organic matter in the three ginseng pools,Bacteroides and Curvularia.Bacteria were positively correlated with each other;the number of bacteria in the water of three kinds of water-raising machines with different operation time had the same trend,and the range of variation of water-raising machines for 6 h,12 h and 18 h was 486-546 genera,512-595 genera and 526-586 genera respectively;PCA analysis showed that the similarity of bacterial communities was 18 h,12h and 6h respectively.
