The mechanism of wheat heterosis is complex,which is one of the difficulties in the field of molecular genetics.In this study,we explored the possible yield-related genes of heterosis through the direction of wheat histone demethylase(HDML)gene family.Histone demethylase is one of the key regulators of biological growth process and an important regulatory mechanism in the process of plant growth and development.In this study,by analyzing the existing genomic data of wheat,a total of46 HDML family genes were obtained,named Ta HDML,which were divided into two subfamilies of Ta LSD and Ta JMJC according to the different conservative domains.According to the chromosome order,they are arranged as Ta LSD1?Ta LSD12,Ta JMJC1?Ta JMJC34.A systematic bioinformatics analysis of 46 Ta HDML family genes revealed that the HDML family gene structure shows complex diversity,with a larger number of exons and introns.Most genes have more than 7 exons,and the gene structure.1 Ta HDML family gene arrangement position on chromosome 1 and chromosome 6 showed high homology,and the other chromosomes had Ta HDML gene distribution.Homologous evolution analysis showed that the wheat HDML gene is highly homologous to rice and Arabidopsis,and is speculated to have similar biological functions.Through the Exp VIP-Wheat Browser(http://wheat-expression.com/)database,the expression data of roots,young ears,leaves and grains during the period of reproductive growth of wheat and the strong dominant two-line hybrid wheat Jingmai6(JM6)Based on the combined transcriptome sequencing data,18 Ta HDML family genes with high differential expression in panicles and grains were screened.Furthermore,the combinations of superparental expression in 40 wheat varieties with different dominant types and the hybrids of Jingmai 6 and Jingmai 21 were used as test materials to verify the expression of 18 highly differentially expressed Ta HDML family genes by real-time quantitative PCR.The results showed that the expression levels of the four wheat HDML family genes in F1 hybrids were significantly higher than those of their parents,showing a super-parental expression pattern.The results ofwheat protoplast subcellular localization showed that Ta JMJC23 was mainly expressed in the nucleus;Ta LSD7,Ta JMJC21,and Ta JMJC25 were all expressed in the nucleus,cytoplasm,and cell membrane.It is speculated that these histone demethylases may play an important histone in the nucleus The function of demethylation regulation may be involved in the regulation network of heterosis gene expression in yield of two-line hybrid wheat.
