Objective:This research pays attention to the rats model of irritable bowel syndrome and investigates the effect of ACC(anterior cingulate cortex)neurons discharge of acupuncture stimulating.Then analyze the molecular mechanism and the law of specificity of acupuncture points from pain producing substance of the target organ.Methods:To establish the rats model with maternal separation,acid enema and colorectum expansion.We evaluated the praxeology of the model rats after the preparation.Rats model with IBS were divided into blank control group,model group,Neiguan group,Zusanli group.The model group was bounded but not given EA,the EA was on Neiguan and Zusanli group.Then test the sensitivity evaluation and emotional behavioral changes with AWR and EPM.Record the changes of the ACC neuron discharge in vivo multi-channel synchronous recording.And then test the rats colon tissue of 5-HT2A receptor expression using immune histochemical methods.Results:1.the influence of the intestinal sensitivity at different acupuncture points on IBS model Contrast with the blank control group,the abdominal constriction latency is shorter(P<0.01),the number is larger of contract wave counting(P<0.01,P<0.01,P<0.05).Contrast with the model group,the abdominal constriction latency is longer(P<0.05,P<0.01)and the contract wave counting is smaller of Neiguan and Zusanli group(P<0.05,P<0.01).2.the influence of the emotional changes at different acupuncture points on IBS model Contrast with the blank control group,the OT%and moving distance is fewer on model group(P<0.05),the OE%and moving distance is fewer on Neiguan group(P<0.05).3.the influence of the 5-HT2A receptor expression at different acupuncture points Contrast with the blank control group,5-HT2A receptor expression increased of model group,Neiguan and Zusanli group(P<0.01).Contrast with the model group,5-HT2A receptor expression decreased of Neiguan and Zusanli group(P<0.01).Contrast with the Neiguan group,5-HT2A receptor expression decreased Zusanli group(P<0.05).4.the influence of the ACC neuron discharge at different acupuncture points on IBS model Unit A:Contrast with the blank control group,frequency of the neuron discharge of model group is increased(P<0.05);Contrast with the model group,frequency of the neuron discharge of Neiguan and Zusanli group is decreased(P<0.01,P<0.05).Unit B:Contrast with the blank control group,frequency of the neuron discharge of model group is decreased(P<0.05);Contrast with the model group,frequency of the neuron discharge of Zusanli group is increased(P<0.05).Conclusion:1.The method of neonatal maternal separation and acid enema can prepare the stable IBS model animal.2.EA on Neiguan and Zusanli can alleviate symptoms of IBS rat model,improve abnormal emotional psychological change in rats.Acupuncture can inhibit the abnormal expression of 5-HT2A receptors,participate in the regulation of pain.3.Effect of acupuncture point is unique.Zusanli is to be specific to the regulation of gastric bowel.We analyzed that target organs peptide material metabolism dysfunction is one of the important mechanisms of irritable bowel syndrome.4.Neiguan could instrain the frequency of the neuron discharge of Unit A,and Zusanli can excite the the frequency of the neuron discharge of Unit B.The connection between accupoints and the frequency of the neuron discharge is to be discussed.
