Objective:To analyze IncRNA expression profiles in MIN6 cells which belong to mice pancreatic ? cells expoused to cytokinesMethods:MIN6 cells were incubated in the presence or absence of cytokines for 24 h,cell apoptosis was detected by flow cytometry and cell viability was assessed by the Cell Counting kit-8;Affymetrix GeneChip Mouse Transcriptome Array 1.0 was used to identify the differently expressed IncRNA and mRNA in MIN6 cells exposed to cytokines compared with normal control group;Gene Ontology(GO)and pathway analyses were used to study the differentially expressed IncRNAs;A IncRNA-mRNA co-expression network was constructed to identify the interactions between the differentially expressed IncRNAs and mRNAs;candidate IncRNAs were verified by real-time quantitative PCR;UCSC and NONCODE database were used to collect the biological information of the differentially expressed IncRNAs.Results:(1)Cytokines can promote MIN6 cells apoptosis and inhibit MIN6 cells viability;(2)723 differently expressed IncRNAs were identified in the cytokine-stimulated group compared with the control group with a set filter fold-change = 2.0,of which 444 upregulated and 279 downregulated.And 2180 differently expressed mRNAs were identified with a set filter fold-change(?)1.5.(3)GO analysis revealed that the up-regulated genes enriched in 462 significant functions,and the down-regulated genes enriched in 312 significant functions.Pathway analysis revealed that the up-regulated genes enriched in 83 significant functions,and the down-regulated genes enriched in 72 significant functions.(4)From the IncRNA-mRNA co-expression network;,we identified the core regulatory IncRNAs and mRNAs.(5)Fifteen IncRNAs were aberrantly expressed in the cytokine-stimulated group compared with the control group.The results of the qRT-PCR were consistent with the data from the microarray.(6)lncRNA NONMMUT042324 which located in mouse chr5,may regulate target gene CXCL10;lncRNANONMMUT036704 which located in mouse chr2,may regulate target gene Lnc2;lncRNANONMMUT034373 which located in mouse chr19,may regulate target gene CD274 antigen.Conclusion:The results reveal that there are differently expressed lncRNAs in MIN6 cells expoused to cytokines compared with the control group.These lncRNAs may play a key role in maintain pancreatic ? cells function.
