Ulcerative colitis(UC)is a kind of chronic,non-specific intestinal inflammatory lesions whose etiology and pathogenesis are unclear,scientists widely believe it is a kind of inflammatory bowel disease that is caused by the interaction of genetic,environmental,and immune factors.In recent years,the incidence is rising in our country,it has a tendency to break out repeatedly and cancer.UC has been listed one of refractory diseases by the world health organization and is one of the hotspot in disease research today.Due to ulcerative colitis is a chronic relapsing intestinal inflammation of unknown etiology and has deferment and recurrent characteristics,so it requires long-term treatmen,but is easy to relapse after drug withdrawal,intolerance of patient and side effects of drugs is one of the key factors which restricts its curative effect.In UC patients need long-term or lifelong chemical drugs or immune inhibitor treatment,side effects and low response rate,looking for a probiotic treatment or auxiliary treatment of UC is a good choice,so probiotics which is good for intestinal mucosa and has immune regulation function has gradually aroused people’s attention.In view of the current problems,in order to achieve the purpose of prevention and mitigation ulcerative colitis and to provide theoretical basis and method for the health care industry and cure of ulcerative colitis,this paper applied Lb.paracasei HD 1.7 which was isolated independently by our laboratory as test strain,at first,we used the method of spray drying to preparate microcapsules of Lb.paracasei HD 1.7,then we applied dextran.sodium sulfate(DSS)to induce mice UC model.BaLb/c mice were randomly divided into 1.the healthy control group(healthy control)2.the Lb.paracasei HD1.7 naked bacteria group(UC+Lb.paracasei HD1.7)3.the microcapsule group(UC+microcapsule)4.the empty microcapsule group(UC+Sterile microcapsule)5.the ulcerative colitis control group(UC+hysiological saline).During the experiment,condition of daily was observed such as mice eating,drinking,hair color,activity and so on,at the same time mice’s droppings shape,stool occult blood,and bloody were observed and mice’s body weight were weighed.7 and 14 days respectively in the experiments,the blood of mice were took with eyeball blood method,we used enzyme-linked immunosorbent method to detect the contents of cytokines and used flow cytometer to test the relative number of T lymphocytes.After that mice were sacrificed using cervical dislocation method,the situation of colon was observed and mersure the length of colon in the mice,mice’s spleen and liver were took out and weighed their weight.The whole colon of mice was sheared,we used enzyme-linked immunosorbent method to detect the content of cytokines and SigA;another part of the colon tissue was used to embedding with paraffin,cut into slices and dyed with HE staining.After that mesenteric lymph nodes and payer patch were sheared,we used flow cytometer to detect the relative quantity of Tlymphocyte.The research results are as follows:(1)Compared with the empty microcapsule group and the ulcerative colitis control group,the microcapsule group and the Lb.paracasei HD1.7 naked bacteria group could significantly slow down downward trend in mice body quality,colon length,DAI and histopathological integral(p<0.05),meanwhile their could significantly improve the the index of spleen and liver(p<0.05),the microcapsule group was the best in the treatment group.There was no significant difference between the empty microcapsule group and the ulcerative colitis control group(p>0.05).(2)Compared with the empty microcapsule group and the ulcerative colitis control group,the microcapsule group and the Lb.paracasei HD 1.7 naked bacteria group could significantly reduce the content of IFN-y and TNF-α in colonic tissue and peripheral blood(p<0.05),the microcapsule group was the best in the treatment group.Althoughthe Lb.paracasei HD 1.7 naked bacteria as well as microcapsule could reduce levels of IL-6 in the colon tissue and peripheral blood,there was no significant difference compared with the empty microcapsule group and the ulcerative colitis control group(p>0.05).Compared with the empty microcapsule group and the ulcerative colitis control group,the content of IL-10 and IL-13 was significantly elevated in the microcapsule group and the Lb.paracasei HD1.7 naked bacteria group(p<0.05),the microcapsule group was the best in the treatment group.(3)Compared with the empty microcapsule group and the ulcerative colitis control group,the content of SIgA was significantly elevated in the microcapsule group and the Lb.paracasei HD 1.7 naked bacteria group(p<0.05),the microcapsule group was the best in the treatment group.The Lb.paracasei HD 1.7 naked bacteria as well as microcapsule could not significantly elevate the relative quantity of CD4+and CD8+ T lymphocyte of the colon tissue and peripheral blood in the microcapsule group and the Lb.paracasei HD 1.7 naked bacteria group compared with the empty microcapsule group and the ulcerative colitis control group(p>0.05).Compared with the empty microcapsule group and the ulcerative colitis control group,the microcapsule group and the Lb.paracasei HD 1.7 naked bacteria group could significantly elevated the relative number of CD4 + and GD8+ T lymphocyte of the payer patch(p<0.05),the microcapsule group was the best in the treatment group.Comprehensive analysis,DSS-induced mice model can simulate ulcerative colitis clinical symptoms,the Lb.paracasei HD 1.7 naked bacteria as well as microcapsule has therapeutic effect on ulcerative colitis in mice,the mechanism of therapeutic effectmay inhibit the inflammatory cytokines(IFN-γ and TNF-α)’ expression and promote anti-inflammatory cytokines(IL-4,IL-10 and IL-13)’ expression and the content of SIgA,meanwhile,the mechanism of therapeutic effect may enhance the relative number of CD4+and CD8+ T lymphocyte.Consequently,the Lb.paracasei HD1.7 naked bacteria as well as microcapsule realize the treatment of UC diseases.
