Objective:By observing Penyankang suppository’s effect on sequelae of pelvic inflammatory disease model rat’s serum pentraxin 3(PTX3),interleukin-8(IL-8),to explore Penyankang suppository’s mechanism in the treatment of sequelae of pelvic inflammatory disease,to provide experimental support for clinical application.Methods:Experimental subjects were SD female rats,90 healthy SPF rats were randomly divided into 3 groups,followed by 10 for normal group,70 for model group,10 for sham-operated group,the model group rats were made model by "phenol mucilage modeling method" to copy sequelae of pelvic inflammatory disease models;14 days after modeling operation,we randomly selected six rats from the model rats group to evaluate if the model was successfully copied;after the model was proved successful,the model rats were randomly divided into Penyankang suppository’s high,middle,low dose group,Kangfuxiaoyan suppository each group 10,model control group 12;the normal group,sham group was the same with the former,a total of seven groups,all rats were treatded continuous 21 days;24 hours after the last treatment,all rats were anatomized to get the uterine and arterial blood saved to be tested;ELISA the PTX3 and EL-8 in blood serum,scope the rat uterus tissue morphology by the light microscope after HE staining.Result:1.Fourteen days after modeling operation,found that model group rats’ uterine tissue were damaged,thickening,endometrial infiltrated with inflammatory cell,interstitial dilatation and congestion,managed SPID pathology.2.Compared with the normal uterine inflammation grading,sham operation group was not significant(P>0.05),model control group scores increased,the difference was statistically significant(P<0.05);compared with the model group grading inflammation of the uterus,Penyangkang suppository high dose group and middle dose group score decreased,the difference was statistically significant(P<0.05),Penyangkang suppository low dose group was not statistically significant(P>0.05);the score difference in each two dose Penyangkang suppository groups was not significant(P>0.05).3.Compared with normal rats serum concentrations of PTX3,sham surgery group was not significant(P>0.05),model group increased,the difference was statistically significant(P<0.05);Compared with model group rats serum PTX3 concentration,each dose Penyangkang suppository group reduced,the difference was statistically significant(P<0.05);the serum PTX3 concentration difference in each two dose Penyangkang suppository groups was not significant(P>0.05)4.Compared with normal group serum IL-8 concentration,the sham group was not significant(P>0.05),control group concentration increased,the difference was statistically significant(P<0.05);compared with the model group IL-8 concentration,each dose group Penyangkang suppository reduce,the difference was statistically significant(P<0.05);the serum IL-8 concentration difference in each two dose Penyangkang suppository groups was not significant(P>0.05).Conclusion:1.Using " Phenol mucilage modeling method " could successfully copy sequelae of pelvic inflammatory disease rat’s model.2.Penyangkang Suppository can reduce the inflammation of the uterus in SPID rats.Serum IL-8 and PTX3 expression elevated in SPID model,after Penyangkang suppository serum IL-8 and PTX3 levels were significantly decreased,indicating the occurrence of SPID of Penyangkang suppository was through infibiting IL-8 and PTX3,thereby reducing inflammation and tissue damage,and promote tissue repair,suppressing the SPID pathological promoting,and play a therapeutic role,so we look IL-8 and PTX3 as Penyangkang suppository treat targets.
