Objective:To discuss the expression characteristic of long non-coding RNA-Co7Rik and to explore its potential role inhepatic gluconeogenesis.Methods:1)Establishing fasting-refeeding model and high fat diet(HFD)feeding model to testthe expression levels of LncRNA-Co7Rik.2)Using different parts of C57BL/6J mice to observe its tissue expression specificity.3)Using UCSC genome browsing tool to analyze the sequencescharacteristic of LncRNA-Co7Rik,and to identify the sequences by nucleic acid electrophoresis.4)Isolating and culturing primary hepatocytes and stimulating with db-cAMP?Forskolin?Dexamethasone and Hydrocortisone to detect the expressionlevels of LncRNA-Co7Rik.5)Using cytoplasm and nucleus separate method to measure the distributionof LncRNA-Co7Rikin cell nucleus or cytoplasm.Results:1)Fasting-refeeding model has shown thatthe relative expression levels of LncRNA-Co7Rik in different times of starvation was 1.00±0.61?2.98±0.87?3.41±0.98?6.73±1.21 and 1.53±0.78(P<0.05).this results shown that theexpression changes of LncRNA-Co7Rik dramatically increased along with the process of starvation,and after refeedingthe expression levcl can returned to normal.2)LncRNA-Co7Rik was up-regulated in HFD induced obesity mice model compared with normal control group(1.00±0.21 vs 2.92±0.63,P<0.01).3)According to RT-PCR.LncRNA-Co7Rik was specific high expression in liver,the expressionwas higher 3-4 times than others(P<0.05).4)The chromosome region of the LncRNA-Co7Rik is Chr8qA1.1,with a length of 1202 bp,and the sequence is found to be consistent with the sequence in the database by nucleic acid electrophoresis,which confirms that the sequence is real.5)In addition,LncRNA-Co7Rik wasactiviated by the signal molecule of gluconeogenesis,which indicatedit might be regulated in the process of gluconeogenesis.6)Subcellular localization suggests LncRNA-Co7Rik mostly distributed in the cell nucleus,indicating it may play an important role in the gene transcription level.Conclusion:LncRNA-Co7Rik isa liver specific IncRNA,which is regulated by glucose signaling and may be involved in the physiological and pathological processes of gluconeogenesis.
