The Expression and Significance of Serum UHRF1 in Patients with Esophageal Squamous Cell CarcinomaObjective:To detect serum UHRF1 levels in patients with esophageal squamous cell carcinoma(ESCC),explore its diagnositic significance for ECSS,analyze its correlation with clinicopathologic features.To analyze the defference of serum UHRF1 pre-and postoperative,and to assess its values of evaluation of operation and dynamic monitoring.Methods:130 preoperative serum samples and corresponding clinicopathological data from patients suffering from esophageal squamous cell carcinoma were collected,of which 62 serum samples postoperative(37 at 1 week postoperative and 25 at 2 week postoperative)and 14 serum samples at both 1 week and 2 weeks postoperative were followed up.67 healthy subjects were served as the control group.The UHRF1 levels were detected by ELISA,Receiver operation characteristic curve(ROC curve)was used to assess the diagnostic efficacy of serum UHRF1,which was compared with tranditional tumor biomarkers.The difference of serum UHRF1 levels in different groups was analyzed by independent t test,paired t test and one-way ANOVA.The relationship between serum UHRF1 levels and clinicopathological parameters were studied with Pearson correlation and Spearman rank correlation.Results:The results showed that the serum UHRF1 levels were significantly higher in patients with esophageal squamous cell carcinoma than healthy controls(t=7.680,P<0.0001).62 serum UHRF1 levels were decreased after esophagectomy(t=5.53,P<0.01)to levels of control group(t=1.622,P>0.05).Successional detection of serum UHRF1 from 14 esophageal squamous cell carcinoma cases showed a decline with statistical difference(F=7.595,P<0.01).Receiver operation characteristic curve demonstrated the sensitivity and specificity of serum UHRF1 for diagnosis was 77.69%and 86.57%respectively,when cutoff was 199.36ng/L.The area under the ROC curve was 0.835(95%CI:0.781?0.890).The diagnostic efficacy of serum UHRF1 for esophageal squamous cell carcinoma was higher than traditional tumor biomarkers including CEA,CA-724,CA-125 and CA-199.In addition,there were positive correlations between serum UHRF1 levels and clinicopathological parameters such as tumor size,lymph node status,invasive depth and pTNM stage,while nagetive correlation was showed between serum UHRF1 levels and differentiation.Conclusion:These results suggest that serum UHRF1 may serve as a novel potential biomarker for diagnosis and dynamic monitoring of esophageal squamous cell carcinoma.Part ? The Impact of Knockdown of UHRF1 on Proliferation and Migration of Esochageal Squamous Cell Carcinoma cell lineObjective:To investigate the Ubiquitin-like with PHD and ring finger domains 1(UHRF1)expression in ECA-109 and KYSE-450 cell line,exploring its effect on cell proliferation and migration when knocked down.Methods:In this study,esochageal squamous cell carcinoma cell line ECA-109 and KYSE-450 was used to investigate the UHRF1 expression and biological function using RNA interference technology.PCR were performed to validate the UHRF1 inhibition effects.Cell proliferation was detected by CCK-8 assay.The migration ability of cell line were analyzed using Transwell chamber tests.Results:In knockdown group,UHRF1 mRNA levels were lower than negative control and blank control cell lines(P<0.05).CCK-8 assay showed that UHRF1 depletion obviously inhibit cell proliferation(P<0.05).UHRF1 depletion leads to the inhibition of cell migration(P<0.0001).Conclusion:Knocking down UHRF1 in esophageal squamous cell carcinoma cell line significantly inhibit cell growth,migration...
