Objective:Exploring content variation regularity of indicative components such as TSG,free anthraquinones and combined anthraquinones contained in Polygonum multiflorum after a nine-time repeat of the steaming and sun-drying process using HPLC;Inspecting factors that influence the processed products and then optimizing the processing technology with orthogonal test so that new processing technological parameter can be provided;Based on what have been done before,we will study on its quality and draft a nine-time repeat of the steaming and sun-drying processed quality standard for Polygonum multiflorum,which can preferably reveal its processing intention;Finally,We will do some primary research on hepatocyte activity processed product make using cell experiment.Which make an account for the reasonability of nine-time repeat of the steaming and sun-drying process.Further,we can provide some experimental evidence for lower-toxicity processing and its clinical medication safety.Methods:1.Study on content variation regularity of indicative components during classical and modern method processing Refering to literature and compared with pharmacopeia method processing,we determinated the indicative component and analyzed its content variations regularity of Polygonum multiflorum after a nine-time repeat of the steaming and sun-drying process using HPLC.2.Study on nine-time repeat of the steaming and sun-drying processing technology.Firstly,we determined the levels and influencing factors beofore carry on the orthogonal experiment.We used indicative components as the inspected index.Then,we optimized an optimum processing condition using the orthogonal test.3.Study on Polygonum multiflorum quality standardAccording to the detection methods that Pharmacopeia of 2015 vertions provided,we detected the character,microscopic identification,thin-layer identification,moisture,total ash,extractum and indicative components content of Polygonum multiflorum with different batchs which were processed by the optimum processing condition so that we could make a preliminary assessment on its quality.Finally,we draught a nine-time repeat of the steaming and sun-drying processed quality standard for Polygonum multiflorum based on the results above.4.Study on hepatocyte activity various processed products of Polygonum multiflorum made Exploring the hepatocyte activity both two processed products made on humans’ L02 liver cells using CCK-8 method and verify the rationality of the nine repeat steaming and sun-drying process.Results:1.Study on content variation regularity of indicative components during classical and modern method processing Of the nine repeat steaming and sun-drying products,the concentration of TSG obviously decreased.While,content of combined and total anthraquinone were overall declined.What’s more,index components of pharmacopeia method processed drew near to the fourth time processed product.2.Optimization on nine-time repeat of the steaming and sun-drying processing technology We chose steaming time(A),sun-drying time(B)and repeat times(C)as factors and set three levels respectively.What’s more,we determined the content of TSG,combined anthraquinone and total anthraquinone as the evaluation index for the orthogonal test.The result showed that A3B1C3 was the optimum processing combination.We determined that to steam four hours,to dry four hours and to repeat for eleven times as the processing condition of Polygonum multiflorum.While,the primary and secondary order for the processing technology influenced factors of Polygonum multiflorum was A>C>B.That was to say,of the three influencing factors,steaming time dominantly influenced the test result,then repeat times,the last was sun-drying time.3.Initially drafting for Polygonum multiflorum quality standard After detecting the character,microscopic identification,thin-layer identification,moisture,total ash,extractum and indicative components content of Polygonum multiflorum with 14 different batchs which were processed by optimum processing condition,we made an quality analysis and got some average value and ranges of its content:average value for moisture was 13.51%with the range from 9.15 to 16.35%;average value for total ash was 2.75%with the range from 2.124.98%;average value for extractum was 5.07%with the range from 0.7710.39%;average value for TSG was 1.43%with the range from 0.493.12%;average value for combined anthraquinone was 0.295 with the range from 0.1470.611%;average value for total anthraquinone was 0.009 with the range from 0.0010.024%.Ultimately,the draft for quality standard established.4.Study on hepatocyte activity various processed products of Polygonum multiflorum made Inhibiting ratio of various processed products on humans’ L02 liver cells was decreased with the processing degree deepen.Conclusion:1.Study on content variation regularity of indicative components Study on content variation regularity of indicative components during classical and modern method processing Processing do influence the content of indicative components.Which indicated that processing has some relationship with toxicity lower.2.Optimizing the nine-time repeat of the steaming and sun-drying processing technology We draft the technological parameter as“steaming for 4 hours,dying for 4hours,and repeatedly for 11 times”.When normalizing the processing index,product quality can be improved.Further,it can also provide some theoretical basis for mass production.3.Initially drafting quality standard for Polygonum multiflorum Polygonum multiflorum processing products should meet its character,microscopic identification and thin-layer identification requirement.And that specifically,content of TSG should not be less than 0.40%;content of combined anthraquinone should not be less than 0.100%;content of total anthraquinone should not be above 0.020%.4.Study on hepatocyte activity various processed products of Polygonum multiflorum made.We found that Polygonum multiflorum did have the objective liver inhibit;Further,there was a positive relationship between the inhibiting ratio and processing degree.In a word,nine-time repeat of the steaming and sun-drying processing method seems more reasonable.
