Objective: To observe the effect of the Hemu-acupuncture therapy on colonic trypatse(Try),proteinase-activated receptor 2(PAR-2),transient receptor potential channel vanilloid subfamily member 1(TPRV1),substance P(SP)and calcitonin gene-related peptidecontents(CGRP),effects in rats with irritable bowel syndrome(IBS)so as to explore its underlying mechanism.Methods: Forty male Sprague Dawley rats were equally randomized into normal group,model group,acupuncture(Hemu-acupuncture)group and medication(Pinaverium bromide,intragastric administration,once daily for 14 d)group.IBS model was established by giving the rat with chronic stressstimulation.Hemu-acupuncture of Shangjuxu point(He point,ST 37),Tianshu point(Mu point,ST25)was given to the rat 30 min,once daily for 14 d.Indicator: Histopathological changes of the colon tissue were displayed by Mast Cell(MC)staining;changes of body weight,determination of colorectal distension(CRD)and abdominal withdrawal reflex(AWR);immunohistochemical staining with Try expression.colonic PAR-2,TRVP1,SP,CGRP expression was detected by Western BlotResults:1.For post-treatment weight changes,model rats showed a significant weight loss,significantly different from the normal and medication groups statistically(P<0.01);2.In terms of CRD volume,differences in abdominal and hip elevation were significantly different between model group and normal group(P<0.01);abdominal elevation was higher in the acupuncture group and medication group than in the model group(P<0.05);and hip elevation was significantly higher than that in the model group(P<0.01).In the behavioral assessment of AWR rats,scores significantly decreased in the model group compared with the acupuncture group and medication roup after treatment(P<0.01).3.There was a significant difference in rat colonic MC count between model group and normal/ acupuncture/ medication group(all P<0.01).and comparison of the acupuncture group with the model group was statistically significant(P<0.05).4.Expression of Try,PAR-2,TRVP1,SP and CGRP were significantly down-regulated in colon tissues in the acupuncture group and the medication group compared with the model group(P<0.01),and abovementioned parameters in colon tissues were statistically significant between acupuncture group and medication group(P<0.05).Conclusion:1.Acupuncture on Hemu-acupuncture reduces hypersensitivity reactions and inflammation of IBS rats as one of effective ways for treating IBS.2.In PAR-2/TRPV1,the expressions of related factors such as PAR-2,TRPV1,SP and CGRP are positively correlated,while the pathogenesis of IBS might be associated with PAR-2/TRPV1.3.Perhaps acupuncture on Hemu-acupuncture reduces SP,CGRP(inflammatory mediator)by inhibiting PAR-2/TRPV1,and regulates hypersensitivity of internal organs.
