With the rapid development of the economy,the style and pace of life has changed in recent years.Breast cancer is one of common cancers which seriously affect women's health.The incidence of breast cancer is increasing year by year,and it shows that the age onset to be younger.But clinical work still needs some new molecular targets.Generally,mammary gland development has been divided into four phases:puberty,pregnancy,lactation and involution.To understand the regularity in mammary gland development process has important academic and practical significance.We can take some measures to achieve the regulation of mammary gland function and to ensure breast healthy.At the same time,this improved the efficiency of milk production.CAMRs family belongs to immunophilin protein family,is a class of highly conserved proteins.They involved in protein folding,immune suppression,and inhibition of T cell activation,signal conditioning and other physiological and biochemical activities.About the role of CAMRs family members in plant and mammalian development is involving in the regulation of hormones in current study.Moreover,CAMRs had been detected the distribution in various tissues and tumors.CAMR1 is an important cytoplasm protein member of the family.Concerning on the function of CAMR1 in mammary gland and breast cancer has not been reported.In this study,we collected mammary gland tissues of normal mice in puberty,pregnancy,lactation,and involution phases and breast tissue in mice which suffering from cancer.According to H&E staining,we observed the morphological changes in normal mammary gland and breast cancer tissues.Using RT-PCR,Western Blot and immunohistochemical analysis,we aimed to investigate the expression level and localization to analyze the role of CAMR1 in normal mice mammary gland development and the process of tumor,so as to further explore the the molecular mechanisms significance during the development of mouse mammary gland and breast cancer.The results analysis showed that:(1)RT-PCR showed that in mRNA levels CAMR1 expressed during various periods of normal mouse mammary development and mice suffering from breast cancer.(2)Histological analysis results:HE staining showed that normal mouse mammary gland development went through four stages of morphological changes.Compared with normal breast tissue,the breast cancer tumors showed significant pathological changes in morphology,including irregular cell shape,disorganized arrangement,varying in different morphology at different stages.PASM and Trichrome staining reveled a phenomenon of fibrosis increasing in breast cancer tumors.Immunohistochemical results showed that,CAMR1 mainly located in the luminal epithelial cell and alveolus cavity in normal mouse mammary gland.However,in breast cancer tumors CAMR1 has been found in the lymphatic site.(3)Western Blot results demonstrate that the expression of CAMR1 proteins during normal mouse mammary gland development significantly changed.Of interest is the observation that with the developmental stages of mamamary gland,CAMR1 proteins level presents a cyclical appearance.It showed that CAMR1 expressed lower levels in puberty,and increased during pregnancy,and reached the highest during lactation.But the expression level of CAMR1 appeared to fall again in the involution phase.In breast cancer tumors,the expression was significantly lower than the amount normal breast tissue.Conclusion:we find out that CAMR1 gene expressed in various periods of normal mammary gland development in normal mice and located in the luminal epithelial cell and alveolus cavity.Interestingly,with the development of the mammary gland,CAMR1 shows cyclical expression changes.It indicates that CAMR1 may participate in the development process of the mammary gland.Also,CAMR1 has been detected in the breast cancer tumors.In addition,the expression level is far below the normal breast tissue.CAMR1 was found in the area of lymphatic vessels.Compared with normal mammary gland,breast cancer tissues showed the feature of the increasing of fibrosis.In all,CAMR1 take part in the normal mammary gland development,it is also closely related to breast cancer.But we need further study to find the specific mechanism of action.
