Polygonatum Rhizoma is the dry rootstocks of Polygonatum kingianum Coll.et Hemsl.、Polygonatum sibiricum Red.、or Polygonatum cyrtonema Hua.of Liliaceae.Because its raw material has the irritation of "irritate man’s throat",it usually needs to be processed after the medicine.The 2015 edition of Chinese Pharmacopoeia recorded that its processing methods are all traditional experience methods,and lack of specific scientific production process parameters.Content:The method under the current Chinese Pharmacopoeia,Indicators in the standard of Pharmacopoeia as an indicator of investigation,Combined with total saponins,EBC and 5-HMF content,optimization of 4 methods of processing for Polygonatum cyrtonema Hua by orthogonal design.To establish a more scientific and modern processing of origin technology suitable for large production,and it provides a basis for its quality control and safety demonstration research.Methods:1.Research on processing of the producing area technology of Polygonatum:(1)study onthe processing technology of Polygonatum(1-10)steam;(2)the wine processing technology of Polygonatum;(3)the steamprocessing technology of Polygonatum;(4)the boiled processing technology of Polygonatum.2.Determination of 5 kinds of processed products of Polygonatum by HPLC method.3.Study on the changes of HPLC-MS components in the processed products of 5 kinds of Polygonatum.Results:1.Research on processing of the producing area technology of Polygonatum:(1)(1-1O)steam best processing technology of Polygonatum:(Steam pressure 0.15 MPa,steam time 1.5h,remove and slices,put into 70℃ drying box to dry 24h,repeat 2 times)for an ideal method of multi steaming;(2)the wine processing technology of Polygonatum:Each 100kg raw Polygonatum,plus 20%alcohol yellow rice wine 20kg,steamed pressure of 0.05MPa,steaming time 1 h,and the drying at 70℃;(3)the steamprocessing technology of Polygonatum:The steam pressure is 0.05MPa,the steaming time is 1 h,and the drying temperature is 70 ℃;(4)the boiled processing technology of Polygonatum:The amount of water is 10 times,the perm time is 3min,and the drying temperature is 70℃.2.Determination of 5 kinds of processed products of Polygonatum by HPLC method,and The best composition of 5-HMF content is the same as that of the fourth,fifth,sixth chapters of the previous article.3.Study on the changes of HPLC-MS components in the processed products of 5 kinds of Polygonatum.Conclusion:In this study,a new processing of 4 kinis of Polygonatum was developed,which made the production process of origin more parameterized and scientific.Meanwhile,the difference of the processed components of 5 new processes was compared with HPLC-MS which provided a basis for quality control and safety demonstration research of Polygonatum.
