Gynostemma pentaphyllum(Thunb.)Makino belonging to the family of Cucurbitaceae,is a kind of Chinese traditional medicinal herb,the whole plant can be used for medicine.Previous studies showed that gypenosides and flavonoids are the active ingredients in Gynostemma pentaphyllum(Thunb.)Makino with the effects of hypolipidemic,blood pressure reduction and antioxidant activity.Basing on the optimization extraction method of gypenosides and flavonoids,this study choose different cultivated strains of Gynostemma(40201,403,404,405,sweet)in Pingli for materials which have been systematically studied on the effect of the growth period,organ,strains on the total saponins content of Gynostemma.This study also can provide a theoretical basis of yield cultivation,breeding,timely harvesting and sustainable development of the resource.The HPLC is used to detect the content of 5 different Saponins in Gynostemma pent.aphyllum(Thunb.)Makino from different province.And different batches standardized extract as experimental materials are used to establish fingerprints and systematicly study their anti-tumor activity.We could effectively provide the standard to control the quality of standard extract via the establishment of fingerprints.The research of anti-tumor effect can provide a theoretical basis for the development of a broader pharmacological activity of the drug.The main results of the experiments are as follows:1.We propose an ultrasonic extraction process via the response surface method to extract gypenosides and flavonoids in Gynostemma.According to Box-Benhnken central composite design principles with four factors and three levels of response surface analysis,the effects of the liquid ratio,extraction time,extraction temperature and ultrasonic power on the yield of gypenosides and flavonoids are discussed.Optimum process parameters are:solid-liquid ratio(A)is 1:40 g/mL,ultrasonic temperature(B)is 45 ?,ultrasonic frequency(C)is 300 W,ultrasonic time(D)is 60 min.This method is reasonable and reliable which can provide a theoretical basis for the rational development and utilization of Gynostemma in future.2.Five Gynostemma cultivars(40201,403,404,405,sweet)in Pingli are set as materials,while 40201 and 403 were approved by the National Variety Approval Committee and are widely cultivated by local farmers.We have systematically studied the effects of growth period,organs,strains influence on saponins content in gynostemma.The experimental results show that 404 and 405 not only have a series of advantages,such as growth better,less plant diseases and insect pests,high output,but also the content of total saponins are higher than the other three strains(P<0.05).The total amount of saponins in the root of the 405 is much higher than other strains(P<0.01),which is the best material as the medicinal resource.3.The HPLC method established by this study has advantages of good separation,good recovery,high precision,low instruments accuracy and high sensitivity.The results obtained from the proposed method show that there is a significant difference in the content of Saponins in Gynostemma pentaphyllum from the different provinces.4.We choose 17 different batches of standardized extracts to establish fingerprints to detect their content of saponin XLIX.12 common peaks have been standarded with good stability,strong absorption and obvious characteristics.The establishment method of fingerprints is stable and feasible,which is coincidence with the requirements of the established fingerprint technology.It can provide the basis for quality of standard extract.5.The anti-tumor effect of standard extract to S180 in mouse is investigated.Five group are set up:high-dose group(150 mg/kg),middle-dose group(100 mg/kg),low-dose group(50 mg/kg),model group?blank model group and positive control group.We found that the tumor weight in three dose group were less than model group(P<0.05),and antitumor effect enhanced with the increase of the dose.During the period of delivery,each dose group mice have a good mental health,good appetite,also weight increase quickly with no obvious side effects.We expect that this experiment can provide theoretical basis on extending development of pharmacological activities for standard extract in future.
