Background:Long noncoding RNAs(IncRNAs),which comprise a class of non-coding RNAs(more than 200 nucleotides)without protein coding potential generated by RNA polymerase ?,many biological processes are connected with IncRNAs,including cell proliferation,differentiation,apoptosis,development and immune responses?As the reports have demonstrated that IncRNAs play critical roles in cancer development and progression,and they have been regarded as biomarkers for diagnostic and prognostic in gastric cancer,prostate cancer,hepatocellular carcinoma,and other cancers.LncRNAs are tumor-specifc and are also very stable in plasma,urine and other easily obtainable body fluid,which indicates that they are good candidate tumor biomarkers.However,conclusions about the diagnostic value of lncRNAs in thyroid cancer are little known.Objective:To detect the expression of lncRNA DOCK9-AS2 in papillary thyroid carcinoma and the effects on the proliferation and migration of papillary thyroid carcinoma cells.Methods:1.Papillary thyroid carcinoma samples and the corresponding adjacent tissues were collected from the First Affiliated Hospital of Nanjing Medical University during June 2016 to June 2017.2.The qRT-PCR was performed to detected the expression of DOCK9-AS2 in the tissues,and the relationship between the expression of DOCK9-AS2 and the clinicopathological data of patients was analyzed.3,The cell transfection technology was conducted to establish the thyroid carcinoma BCPAP cell downregulating DOCK9-AS2 stably.4.The CCK8 was carried out to investigate the influence of DOCK9-AS2 on the proliferation of the BCPAP cell.5.The wound-healing assays were used to dectect the influence of DOCK9-AS2 on the migration of the BCPAP cell.Results:1.The expression of DOCK9-AS2 in papillary thyroid carcinoma tissues was significantly higher than that of adjacent tissues(P<0.05).2.Expression of DOCK9-AS2 in PTC tissues is related to lymph node metastasis and extraglandular invasion(P<0.05).3.Downregulation of DOCK9-AS2 could suppress the proliferation and invasion of the BCPAP cell(P<0.05).Conclusion:The expression of DOCK9-AS2 in papillary thyroid carcinoma was significantly increased,and Downregulating DOCK9-AS2 could inhibit the proliferation and migration of papillary thyroid carcinoma.
