Objective:To study the effects of bone marrow mesenchymal stem cells(BMSCs)transplantation at Fengfu acupoint on the proliferation and differentiation ofendogenous neural stem cells in hippocampus of dementia rats,and to explore new techniques and methods for the treatment of Alzheimer's disease(AD).Methods:Ten AD rats were randomly selected from 50 AD rats as normal control group.The AD model was established by lateral ventricular perfusion of Abeta 1-42 oligomer.Then they were randomly divided into model group,Fengfu acupoint group,Zusanli acupoint group and caudal vein group,with 10 rats in each group.After the successful establishment of the model,the corresponding intervention was carried out.BMSCs was transplanted at Fengfu acupoint group and Zusanli acupoint group,and the same amount of BMSCs was transplanted in tail vein group.The model group was injected with the same amount of saline at Fengfu acupoint,while the normal control groupwas taken as the blank control group.Four weeks later,the learning and memory abilities of rats were tested by water maze test,and the expression of nestin~+,beta-tubulin-III~+in hippocampus was detected by immunofluorescence to complete the cell count.Results:1.Water maze detection model:The escape latency(EL)of the model group waslonger than that of the control group(p<0.01),and the times of crossing the platform were significantly reduced(p<0.001).2.Test results of water maze after intervention:Compared with the model group,the EL values of Fengfu acupoint group and Zusanli acupoint group were notsignificantly shortened after BMSCs acupoint transplantation(p>0.05),and thenumber of crossing plateau was not significantly increased(p>0.05),while the EL values of caudal vein group were significantly decreased(p<0.001),and the number of crossing plateau was significantly increased(p<0.001);compared with the normal control group,the EL values of caudal vein group were significantly prolonged(p<0.01),and the number of crossing The number of times decreased(p<0.05).3.Immunofluorescence test results:3.1 The results of Nestin~+cells countCompared with the model group,the number of Nestin~+cells in Fengfu acupoint group(p<0.05),caudal vein group(p<0.01),and Nestin~+cells in Zusanli group increased significantly,while the number of Nestin~+cells in Zusanli group did notincrease significantly(p>0.05);compared with the tail vein,there was no significant difference in the number of Nestin~+cells in Fengfu acupoint group(p>0.05).3.2 The results of beta-Tubulin-III~+cells countCompared with the model group,the number of beta-Tubulin-III~+cells in Fengfu acupoint group and Zusanli group did not increase significantly(p>0.05),while the number of beta-Tubulin-III~+cells in caudal vein group increased significantly(p<0.01).Conclusion:BMSCs transplantation at Fengfu acupoint can induce the proliferation of endogenous neural stem cells in the hippocampus of AD rats,but it has not been proved that it can differentiate into neurons.
