Articular cartilage injury is a common disease in the movement,due to the particularitofarticular cartilage,the lack of blood vessel and nerve tissue,lead to their repair co.,LTD.,articular cartilage for transparent cartilage,articular cartilage injury symptoms caused by has a lot of,mainly show the relevant section of the swelling,pain,swelling,and accelerated the degeneration of articular cartilage.Joint damage caused by the damage,we usually call it Osteoarthritis(Osteoarthritis,OA),and in the field of medicine for the treatment of articular cartilage injury,so damaged cartilage repair has been the research hotspot in the field of orthopedics,in clinical treatment of joint damage is more conservative,the symptoms of joint damage can only be for a period of time can only alleviate cannot be totally recovered,traditional treatment is to remove necrotic tissue or corresponding cartilage transplantation,the traditional repair tissue is usually fibrocartilage.With the development of life science and engineering science,stem cells have provided new methods to treat cartilage injury in tissue engineering.1.Choose mechanical separation on acquisition methods of duck cartilage and type,? collagenase digestion method with the method of separation of duck embryo tibial cartilage of cartilage cells,cartilage cells by extend the separation and purification,the cultivation to 18 generation,again after the freeze on the sample using cell activity is higher,through a series of technical means(rt-pcr,flow cytometry inspection car immunofluorescence test,surface specific markers)identifies its characteristics of the cartilage cells.2.multifunctional differentiation potential of chondrogenic stem cells.In the aspect of growth characteristics of chondrogenic stem cells,the experiment of cell cloning ability formation was designed and the growth curve was plotted statistically.In order to verify the multifunctional differentiation potential of chondrogenic stem cells,chondrogenic stem cells were differentiated into adipoblast cells,osteoblasts,chondroblasts and myoblasts,and related genes were detected by rt-pcr technology and immunohistochemical technology after differentiation.The results showed that the chondrogenic stem cells were multifunctional and differentiated into target cells,which were consistent with the characteristics of target cells.3.The use of papain,collagenase knee joint cartilage injury in mice model was ?established,to damage the organizational structure of paraffin embedding,pathological dyeing after using the principle of histology,observing damaged cartilage damage structure.When design the cartilage cells involved in sports injury,with CM-Dil cells cultured cartilage satellite cells tracer tag and injected into the knee joint cartilage injury in mice,set seven time gradient,damaged part in each gradient are cartilage tissue and blood do paraffin pathology biopsy and frozen section,observe change of tissue morphology,cell tracer.The results show that cartilage stem cells can be used to repair cartilage damage.This study confirmed that the duck cartilage stem cells to treat cartilage injury in mice,the successful separation of cartilage cells in vitro,and the rapid growth and multi-function differentiation potential of stem cells for cartilage is verified,the use of stem cells in cartilage repair cartilage damage in mice,providing theoretical basis for stem cells to promote tissue regeneration to repair,to provide experimental basis for the clinical application of stem cells in the treatment of patients with sports injury.
