Objective:by studying the life of fetal pill for recurrent abortion decidual rats water channel protein 2(AQP2)and water channel protein 5(AQP5)positioning differences in expression and distribution of the situation,from the level of protein molecules to clarify life of fetal pill could fetus mechanism,looking for a life of tires pill fetus targets,and the prevention and treatment of recurrent abortion for traditional Chinese medicine to provide new train of thought and experimental basis,improve the level of prevention and treatment of spontaneous abortion.Methods:1.Modeling:after the successful mating of SPF grade SD rats in cage with a ratio of 2:1 male and female,the female pregnant SD rats were randomly assigned to set up shoutai pill group with high,medium and low dose,distrasone western medicine control group,model group and normal control group.The recurrent abortion model was established by gavage with hydroxyurea combined with mifepristone tablets.2.Dosage:since the first day of pregnancy(found female vaginal suppository or vaginal smear)on the day of a large number of sperm began to fill the stomach,in addition to the normal control group,both groups pregnant rats from day 1 to 9 days pregnancy every afternoon to hydroxyurea lavage(450mg/kg),2ml/d saline solution,pregnancy made on the morning of 10 days(RU)486 mifepristone lavage(4.0mg/kg),2ml/d saline solution;In addition,in the morning of each day,the high-dose group(2.0mg/kg),the medium-dose group(1.0mg/kg),the low-dose group(0.5mg/kg)and the group(3.02mg/kg)were given corresponding therapeutic drugs,2m L/d of normal saline was dissolved,and the model group and the normal control group were given normal saline in the morning,2m L/d.After 11 days of gestation,the rats were put to death by anesthesia,and the decidua of the uterus were isolated.3.Detection:the localization and quantitative expression of aquaporin(AQP2)and aquaporin(AQP5)were detected by immunohistochemistry.The expression sites of AQP2 and AQP5 in cells were observed after images were collected under the microscope.Meanwhile,IPP(image-pro)6.0 software was used to analyze the images,measure and calculate the IOD/Area,and the IOD/Area value was taken as a semi-quantitative statistical analysis parameter.Results:1.AQP2 and AQP5 are expressed in the decidua of SD rats,and in the cytoplasm and membrane.2.Compared with the normal pregnancy group,the expression of AQP2 and AQP5 in the model group was significantly reduced,and the difference was statistically significant(P?0.01).Compared with the model group,the expressions of AQP2 and AQP5 were increased in the high,medium and low dose shoutai pill group and distrerone western medicine group,and the differences were statistically significant(P?0.01).The expression of AQP2 and AQP5 in the high-dose shoutai pill group was higher than that in the medium-dose shoutai pill group,the low-dose shoutai pill group,and the disoprogesterone group,with statistically significant differences(P?0.01).There was no significant difference between the ditreprogesterone group and the shoutai pill medium dose group(P>0.05).Conclusion:1.The expression sites of AQP2 and AQP5 in the decidual membrane and cytoplasm of rats with recurrent abortion were found.2.In the decidua of rats with recurrent abortion,the expressions of AQP2 and AQP5 were down-regulated.shoutai pill may be one of the mechanisms of placenta placenta by up-regulating the expression of AQP2 and AQP5 in the decidua of rats with recurrent abortion.3.Aquaporin AQP2 and AQP5 may be the functional targets of shoutai pill and the molecular basis of "kidney water main".
