ObjectiveThe meridian theory is one of the core theories of traditional Chinese medicine.The meridian symptom is an important part of the meridian theory,which directly guides clinical dialectics and provides a basis for the treatment of diseases.This study intends to observe the effect of the behavior and the organs of the rats by ligating on the low hydraulic resistance channel(LHRC)which establishing a meridian blockage model.Whether there will be related meridians syndrome,and whether the effects of obstruction of different meridians are different.Attemping to establish a illness syndrom observation method for the pathology of the meridian,and provide experimental research basis for pathological models and evaluation methods for meridian disease research.Method1 Experiment 1--The effect of behavior and organs by ligating on Stomach and Kidney Meridian in rats41 male SD rats were selected by the open field experiment with similar scores from 60 male SD rats,which were divided into the control group(CG1)=10,Non-ligation group(NG1)=10,Stomach meridian ligation group(SMG)=10 and Kidney meridian ligation group(KIG)=11.The sugar preference experiment was performed before modeling.The ligation method of rats:The CG1 was given anesthesia and removed abdominal hair.The other three groups were used a meridian tester to measure the low electric resistance line(LERL)along the stomach meridian,the kidney meridian and the area of the back without meridians,and ligating on three groups’ part.The overall condition and body weight of the rats were observed.The open field experiment was observed once a week for five times.The sugar preference test was performed again in the sixth week after modeling.The gastric residual rate and intestinal propulsion rate were measured in the seventh week.Besides,the organs were dissected and made morphological observation,the organ coefficients were calculated in the last week.2 Experiment 2--Effects of learning and memory and brain coefficient by ligation on Du Meridian in rats.30 male SD rats were randomly divided into the control group(CG2),Non-meridian ligation group(NG2)and the Du Meridian ligation group(DG),with 10 rats in each group.The CG2 was given anesthesia and removed abdominal hair.The NG2 was ligated on the area without meridians.Use the Meridian tester to find out the corresponding parts of the Du Meridian.After the ligation,the changes in body weight were observed and the Mirrors water maze experiment was performed in the eighth week.After that,the brain was weighed and the organ coefficient was calculated.Result1 Experiment 11.1 Overall condition:Before and after the experiment,the CG1 and the NG1 had normal hair,the mental state was good and the reaction was sensitive.With the experimental time increasing,the activity and the irritability were reduced.The stool of the rats in the STG were dry and granular,rats were active and the other overall condition was the same as the CG1.In the KIG,the hair was loose and dark,the activity was less,liking to be surrounded,thin and irritating.1.2 Body weight:The weight in the KIG was lower than that of the CG1 and NG1 at the second week,the fourth week and the sixth week(P<0.05 or P<0.01).1.3 Open field test(OFT):In behavior score aspect,compared with the CG1,the STG was decreased significantly in the first week(P<0.05)and increased significantly in the fourth and fifth weeks(P<0.05).The KIG was decreased significantly in the first week(P<0.01)and decreased significantly from the second to the fifth weeks(P<0.05).Compared with the NG1,the STG showed a significant decrease in the first week(P<0.05)and a significant increase at the fifth week(P<0.05).The KIG was decreased significantly every week after blocking meridians(P<0.05 or P<0.01).In the residence time of the central area aspect,Compared with the NG1,a significantly decreased were showed in the STG in the first week after the establishment of the model(P<0.05).1.4 Sugar preference test(SPT):Compared with the CG1 and the NG1,the sugar preference value of the STG was significantly increased(P<0.01,P<0.01).1.5 Gastric residual rate and small intestine propulsion rate:The small intestinal propulsion rate of the LSTMG was significantly higher than that of the CG1 and the NG1(P<0.05,P<0.05).1.6 Organ coefficient:Compared with CGI and NG1,the cardiac and renal coefficient of the STG were increased significantly(P<0.01,P<0.01).Compared with CGI and NG1,the epididymis coefficient of the KIG was decreased significantly(P<0.01,P<0.05).The adrenal coefficient of the KIG was increased significantly(P<0.05,P<0.05)and prostate coefficient was decreased significantly(P<0.05,P<0.01).1.7 Organ observation:There were 4 cases of abnormal heart size with atrial hypertrophy,coronary artery engorgement,apical became blunt circles in the STG.There were 5 cases of lung color changes in the STG and KIG,4 cases and 3 cases of color change in the CGI and NG1 respectively,mainly manifested as hyperemia.In the KIG,5 cases of dark color were seen in the liver.In the STG,6 cases of surface abnormalities were seen and the granules like rice size were observed in kidney,3 cases had morphological changes and abnormal surface which most of them were swollen,containing a lot of turbid liquid,and the kidney’s membrane were difficult to peel off.The liver,testicles,and intestines of the STG were occasionally abnormal,and the heart,spleen,stomach,testis,epididymis,prostate,seminal vesicle and intestine were abnormal in the KIG.In addition,there was a hematoma near the abdominal aorta in the STG.1.8 Morphological results:The morphology of the heart,lung,liver,kidney,stomach,testis and epididymis of CGI and NGlwere basically normal.The cardiomyocytes of STG were disordered and slender,the muscle fiber gap was widened,multiple and segmental nuclear pyknosis and necrosis occured.Liver tissue bile duct epithelial cells proliferate with a large number of inflammatory lymphocytes infiltrated.The kidney can be seen in the distal convoluted tubule nucleus pyknosis and vacuolization,mesangial cell hyperplasia,dense plaque necrosis and dissolution.The epithelial layer,lamina propria and mucosal muscle layer of gastric mucosa were unclear and structurally disordered.The testicular tissue spermatogenic cells are loosely arranged,the number of cell layers is reduced,and there are spermatogenic cells falling off in a few seminiferous tubules,and testicular interstitial edema.The epididymis tube is atrophied and deformed,and some of the sperm in the lumen breaks in the head and tail,and is granular.The myocardial cells in KIG were disordered,atrophied,nuclear pyknosis,necrosis,and obvious interstitial edema.Hepatocytes swell,the nucleus multi-divided into a binuclear,bile duct hyperplasia,scattered in the nucleus pyknosis and coagulative necrosis,cytoplasm red staining,unclear liver cell boundaries.Glomerular swelling,epithelial cell swelling and necrosis,tubule cavity disappearance,interstitial vascular congestion,inflammatory cell infiltration.The gastric mucosa epithelium was largely shed and mildly bruised.The shape of the seminiferous tubule is elliptical,the spermatogenic cells are arranged loosely,the number of cell layers is reduced,the vacuolar-like degeneration,and the wall of the seminiferous tubule is thinned.The structure of the epididymis tube was normal,but most of the sperm were broken at the head and tail.They were scattered in the lumen and had abnormal pigmentation.2 Experiment 22.1 Results from the test of gaining concealed platform:the escape latency of mice in each showed varying extends of declining with the increase of training time.Compared with CG2,the escape latency of DG was significantly increased from the 3rd day to the 6th day of training(P<0.01).Compared with NG2,the escape latency of DG was significantly increased on the 2rd day,5th day and 6th day of training(P<0.01,P<0.01,P<0.01).There was no significant difference between the CG2 and the NG2(P>0.05).2.2 Results of spatial trial:The proportion of target quadrants and time of DG was significantly decreased than that of the CG2(P<0.01,P<0.01).The number of crossing the original platform position of DG was the lowest in the three groups,which was significantly decreased than that in the CG2(P<0.05).2.3 Weight:The weight of the DG was significantly decreased than that of the CG2 at the 4th week after ligation.There was no significant difference between the other groups(P>0.05).2.4 Brain coefficient:There was no significant difference between the three groups(P>0.05).Conclusion1 Overall conditions such as body weight,coat color,and mental status indicate that the growth and development of rats is most obvious when ligation on the LHRC in the KIG.Rats in the STG also had changes in stool and activity.It shows that the ligation of the meridians can be associated with a meridian-specific change.2 In the behavioral experiment,the emotion of the STG and KIG were observed by the OFT and the SPT.Ligation on the stomach and kidney meridian LHRC showed the meridian manifestations related to the stomach and kidney meridian.The performance of the STG showed similar evidence of excess syndrome of stomach meridian,and the KIG showed symptoms similar to the deficiency of kidney meridian.The Mirrors water maze was used to observe the decline of spatial learning and memory ability after ligation on Du Meridian.3 In the observation of organs,ligation on the LHRC of the stomach meridian can cause gastrointestinal hyperactivity,affecting the growth of heart and kidney,and change the morphological structure of heart,liver,kidney,stomach,testis and epididymis.The rats of KIG affected the organ coefficients of the adrenal gland,epididymis and prostate,and also affected the morphology of the heart,lung,liver,kidney,stomach,testis and epididymis.Blocking the meridian is closely related to multiple organs.4 Ligation on the different LHRC can produce meridians symptom and organ changes,which proves that blockage the LHRC is one of the reason of meridian symptom,providing new ideas for meridian research and method.
