Objective: MiR-590-3p has been reported to be a tumor suppressor in various types of cancer.However,the role of miR-590-3p in papillary thyroid cancer(PTC)is not well understood currently.In the present study,we have explored the function of miR-590-3p in PTC.Methods: We examined the miR-590-3p expression in human PTC tissues and cell lines by RT-PCR.Luciferase reporter assays was used to validate the target of miR-590-3p.CCK-8,Edu,wound healing,transwell,apotosis assays and western blot were performed to analyze PTC cell function.Results: We found that the expression of miR-590-3p was decreased in PTC tissues and cell lines.Compared with the normal control,knockdown of miR-590-3p promoted cell proliferation,migration,invasion and inhibited cell apoptosis in PTC cells(P<0.01).However,overexpression of miR-590-3p inhibited cell proliferation,migration,invasion and promoted cell apoptosis in PTC cells(P<0.01).High-mobility group AT-hook 2(HMGA2)was identified as a major target of miR-590-3p.Moreover,miR-590-3p inhibitor promoted cell proferation,migration and invasion ability and these effects were rescued by HMGA2.Whearse,knockdown the suppression of cell proferation,migration and invasion caused by miR-590-3p mimics is relieved by overexpression of HMGA2.Conclusion: MiR-590-3p acts as a tumor suppressor role in PTC cell proliferation,migration and invasion via targeting HMGA2,and may be a therapeutic target for PTC.. |
