Purpose:Using mulberry Zhike granules to treat cough variant asthma(CVA)guinea pig model,To observe the signs of guinea pigs,the number of coughs,histopathological changes of lungs,the content changes of HIS and LTB4 in serum and BALF,and the changes of IL-4 and IFN-? level in lung tissue,explore its pharmacodynamics and mechanism of action,provide powerful theoretical support and experimental basis for clinical use.Material and method:Through the ovalbumin sensitization and atomization inhalation cough stimulated,replicate cough variant asthma in young guinea pig model,48 guinea pigs were randomly divided into: normal control group,model control group,western medicine control group,low dose group of mulberry cough granules,middle dose group of mulberry cough granules,high dose group of mulberry cough granules,total 6 Groups,8 in each group.The general state of guinea pigs was observed.The number of coughs stimulated by capsaicin,the histopathological changes of guinea pig lungs,the contents of HA and LTB4 in serum and BALF and the levels of IL-4 and IFN-? Variety.To explore the pharmacodynamic effects of mulberry cough granules and its mechanism of action on cough variant asthma.Results: 1.Observation of guinea pig physical signs: The normal control group had the best state,and the model control group had the worst state.All the other groups had improved compared with the model group,However,the low-dose Mulberry cough treatment group was not as good as the western medicine control group and the middle and high-dose group of Morinda mulberry.2.Counting of cough times: The number of coughs in the model group was significantly more than that in other groups,and the difference was statistically significant(P<0.05);Comparison of the number of coughs between the western medicine group,the middle and high-dose group of mulberry cough granules,The difference was not statistically significant(P>0.05).However,there was a significant difference in the number of coughs in the low-dose group and the western medicine group(P<0.05).There was no significant difference in the high dose group of mulberry Zhike particles(P>0.05).3Lung pathology: There was no obvious pathological change in the lung tissue of guinea pigs in the normal group;the pathological changes in the model group were the most obvious;Each drug group followed.4.ELISA results for HA and LTB4: HA and LTB4 levels in serum and BALF in model group were significantly higher than those in other groups,The difference was statistically significant(P <0.05);HA and LTB4 content was no significant difference in the Western medicine group?the mulberry Zhike particles middle-dose group,and the mulberry Zhike particles high-dose group.However,there were significant differences in the content of HA and LTB4 between the mulberry Zhike particles in low dose group and the western medicine group.The content of HA and LTB4 in the mulberry Zhike particles in low dose group,the mulberry Zhike particles dose group,the mulberry Zhike particles high-dose group were significantly different.5.Immunohistochemical results of IL-4 and IFN-?: The model group compared with other groups,the differences were statistically significant(P<0.01).Compared with the western medicine group,the difference between the mulberry Zhike particles in low dose group was statistically significant(P<0.01).Compared with the western medicine group,the difference in the mulberry Zhike particles dose group,and the mulberry Zhike particles high-dose group was no statistically significant(P>0.05).Conclusion: 1.The mulberry Zhike granules can effectively improve the general state of cough variant asthma guinea pigs,cough symptoms,reduce lung inflammation and reduce the degree of chronic airway inflammation in guinea pig model.2.The mulberry Zhike granules can significantly reduce the content of HA and LTB4 in the serum and BALF of the cough variant asthmatic model guinea pigs,and its mechanism of action in the treatment of cough variant asthma may be related to the regulation of the levels of HA and LTB4 involved in immune inflammation.3.The mulberry Zhike granules can significantly reduce the cough variant asthma model of guinea pig lung IL-4 content,increase the content of IFN-?,can effectively inhibit the release of inflammatory cytokines in immunological mechanisms have a regulatory role.
