Purpose:To investigate the effect of Bushenjiannao electro acupuncture therapy on the rapid aging of SAMP8 in the cortex neurons,A beta and A formation of beta IDE degrading enzyme.To observe the changes of kidney and brain electro acupuncture therapy on the fast aging SAMP8 mice nerve pathology,explore the kidney and brain electro acupuncture therapy in the treatment of AD part mechanism.Material and method: 8 month old male fast aging 18 SAMP8 mice were randomly divided into model group,EA group,memantine,western medicine group(n=6).6 SAMR1 mice with normal age of the same month age were used as negative control group.8 month old male SAMR1 and SAMP8 were electroacupunctureed at "Baihui" and "Shenshu" and "Taixi" point,consecutive 8W.Using Nissl staining,immunohistochemical method and Western blot method,the distribution of neurons and the expression of A beta and IDE were observe in the brain tissue of mouse cortical neurons.Results: 1.Compared with normal aging SAMR1 mice,the cortical neurons of SAMP8 mice in model group were smaller,and more cells lost their integrity.Nissl bodies were lighter and less in number,and the total area and integral optical density of Nissl bodies decreased.Compared with the model group,the volume of cortical neurons in SAMP8 mice increased,and Nissl bodies were denser without obvious vacuolation and vacuolar degeneration.The average area and integral optical density of Nissl bodies increased.2.Compared with the normal aging SAMR1 mice,the level of A beta in the cerebral cortex of the model group SAMP8 mice increased significantly.Compared with the model group,the level of A beta in the cortical brain tissue of SAMP8 mice decreased after the treatment of electroacupuncture and the treatment of the western medicine.3.Compared with the normal aging SAMR1 mice,the expression of IDE in the cerebral cortex of the model group SAMP8 mice decreased significantly.Compared with the model group,the expression of IDE in the cortical brain tissue of SAMP8 mice increased after the treatment of electroacupuncture and the treatment of the western medicine.Conclusion: Effect of Bushenjiannao acupuncture therapy can promote the expression of SAMP8 IDE in the mouse brain.Thereby inhibiting the formation of SAMP8 in the brain of mice A beta,and then improve the morphology and function of neuron SAMP8 in the mouse brain.The results showed that Bushenjiannao electroacupuncture based on “kidney and brain” theory has good effect on the prevention of AD.
