Objective: It is clear that the inflammatory factor and intestinal mucous membrane proteins are involved in UC main pathogenic process.So how to balance disorder and intestinal mucosa inflammation factor protein expression has become a new way of thinking,which has placed an important role in the treatment of UC.Moxibustion as a kind of not directly stimulate intestinal mucosa,but through multi-channel,multi-target,multi-level,comprehensively regulates the function of various parts of the body to achieve the prevention and treatment of repair and protection of intestinal mucosa.It has broad application and development prospects.Inflammatory factors and intestinal mucosal proteins are involved in the main pathogenic process of UC.Balancing the disturbance of inflammatory factors and the expression of intestinal mucosal proteins has become a new way to treat UC.Based on this,the model of ulcerative colitis in rats was established by establishing a rat model of ulcerative colitis.We observe the effect of moxibustion Guanyuan and Zusanli point on the expression of MUC2 and EGFR in intestinal mucosa of rats with ulcerative colitis.In order to reveal the immunological mechanism of moxibustion in the treatment of UC and the mechanism of moxibustion in the aspects of morphology,pathophysiology and immunology.Materials and methods: Fifty healthy adult SD rats were randomly divided into normal group,model group,western medicine group,moxibustion treatment group,and moxibustion combined with western medicine treatment group.DSS free drinking method was used to pre-test 4%DSS solution,and each rat was fed with sodium dextran sulfate(DSS)solution according to drinking 25 ml solution.The new DSS solution was replaced every day.After drinking for 7 days,normal drinking water was given.At the end of the experiment,blood was collected from abdominal aorta of anesthetized rats,and the diseased colon tissue was intercepted.The expression of MUC2 and EGFR protein was detected by immunofluorescence staining,and the expression of MUC2,EGFR gene in colon tissue was detected by RT-PCR method.Results:1.After the therapy of moxibustion and western medicine,the rats' colon histomorphology(immunofluorescence staining)were improved than before,especially in the moxibustion combined western medicine group.2.The rats' DAI score and CMDI score: Compared with normal group,the DAI score and CMDI score in model group increased.After the treatment of the moxibustion,western medicine,and the moxibustion combined with western medicine,the DAI score and CMDI score decreased gradually and approached to the normal level gradually.3.After treatment,the Immunofluorescence expression of MUC2 and EGFR protein increased significantly,especially in moxibustion combined with western medicine group.4.Compared with the model group,the contents of MUC2 m RNA and EGFR m RNA in three treatment groups were increased by RT-PCR(P < 0 05),but there was no significant difference between moxibustion group and western medicine group(P > 0 05).The results showed that the curative effect of moxibustion was similar to that of western medicine,especially in moxibustion combined with western medicine group.Conclusion:1.Intestinal mucosal barrier damage plays an important role in the pathogenesis of UC.2.MUC2 protein and epidermal growth factor EGFR are the main factors of intestinal mucosal barrier damage in UC.3.Moxibustion at Guanyuan point and Zusanli(Zusanli point)can interfere with UC by raising the levels of MUC2 and EGFR.
