Purpose:To study the inhibitory effects of curcumin on proliferation of primary cells derived from patients with high-grade glioma testing different concentrations by collagen gel droplet embedded culture drug sensitivity test?CD-DST?,and if low concentration curcumin can make Temozolomide?TMZ?more sensitive.Material and method:The 59 specimens used for the test were obtained by surgical resection and all primary high-grade gliomas?WHO III-VI?certified by pathologic diagnosis,none of the patients have ever underwent neoadjuvant chemotherapy or radiotherapy before the surgeries.The specimens were made into collagen-cell mixture.Three droplets of the mixture were placed in each well of a 6-well multi-plate and allowed to gel at 37?in a CO2 incubator.Then TMZ?different concentrations of curcumin were added respectly,and combined low concentration of curcumin with TMZ comparing with DMSO as comparison.Washed the drugs after 24 hours and incubated in serum-free culture medium for the subsequent 5 days.Neutral red was added to each well,the color values of tumor cell/control cell were tested of each drop by Primage,to observe the effect of different concentrations of drugs and different drugs on the proliferation of glioma cells.Results:A total of 52 data were enrolled in 59 specimens.Curcumin has inhibitory effect on human primary glioma cells,its inhibitory effect is concentration dependent.The in vitro sensitivity was expressed as the T/C ratio?%?,a T/C?50%to an anticancer drug was regarded highly sensitive,a 50%<T/C?60%was regarded moderately sensitive,T/C>60%was regarded low sensitive.In the 52 specimens,fifteen of them were highly sensitive to curcumin with a concentration of 20?mol/L,twelve were moderately sensitive and twenty-five were low sensitive;Four specimens were highly sensitive to TMZ with a concentration of 10.3?mol/L,thirteen were moderately sensitive and thirty-five were low sensitive;twenty-two specimens were highly sensitive to TMZ with a concentration of 10.3?mol/L combined with curcumin with a concentration of 20?mol/L,sixteen were moderately sensitive and fourteen were low sensitive.Conclusions:1.The collagen gel droplet embedded culture drug sensitivity test can be used for curcumin.2.Curcumin has inhibitory effect on primary high-grade glioma cells,its inhibitory effect is concentration dependengt.3.Curcumin with a low concentration?20?mol/L?can be involved in sensitizing TMZ.
