Purpose: To establish the model of acute gouty arthritis(AGA)rat in sodium uric acid crystallization(MSUC)joint endovascular injection,and to explore the effects of Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)on IL-1β,TNF-α inflammatory factor and TLRs/MyD88 signaling pathway in AGA rats,from pathological tissue,cell and molecular levels.This paper discusses the inhibitory effect of Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)on the inflammatory response of AGA,and partly expounds the effect of Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)on improving symptoms,and provides scientific basis for the clinical treatment of AGA.Material and method: 1.SPF SD Rats 48 randomly divided into normal group(A),model group(B),Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)low dose Group(C),Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)in the dose Group(D),Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)high dose Group(E),An Kang Xin Group(F).8 each in each group.Experiment after 1 weeks of feeding.Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)group and An Kang Xin group to take the Gastric way,Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)1 times/d,of which low dose group 208.33mg/kg/d,medium dose group 416.67/mg/kg/d,high dose group 625mg/kg/d,An Kang Xin 12.5mg/kg/d.The remaining groups were filled with saline and the course of treatment was 5 days.On the 3rd day of drug use,an acute gouty arthritis model was established by injecting sodium uric acid solution in the anterior tibial cavity of the right ankle,and the normal group was injected with the same volume of saline.2.The rats were anesthetized and executed 1 hours after the last gastric lavage,and the materials were taken.The pathological changes of synovial tissue in rat joints were observed by he staining method.The circumference of the right posterior ankle of 2h,6h,20 h and 72 h AGA rats was measured by ruler method.Determination of IL-1β and TNF-α in vein blood of rats by double antibody sandwich Elisa.The expression of TLR2,TLR4 and MyD88 in synovial tissue of rat joint was determined by immunohistochemical method.Results: 1.Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)had a certain inhibitory effect on ankle swelling in AGA rats.2.Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)has a certain inhibitory effect on IL-1β and TNF-α in serum of acute gouty arthritis rats.3.Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)inhibited the upstream protein TLR2,TLR4 and MyD88 of TLRs/MyD88 pathway in the ankle synovial tissue of AGA rats.Conclusion: 1.Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)can reduce the swelling index of ankle joint in AGA rats,reduce the content of inflammatory factor IL-1β and TNF-α in serum,and show that traditional Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)has anti-inflammatory and detumescent effect.2.Chinese herbal Medicine Lai Fu(Northeast female carrot concentrated ultrafine powder)had obvious inhibitory effect on the expression of upstream protein TLR2,TLR4 and MyD88 of TLRs/MyD88 pathway in the ankle synovial tissue of AGA rats,suggesting that its mechanism might be achieved by inhibiting the TLRs/MyD88 pathway.
